Objective: To detect the plasma proteomic patterns using SELDI-TOF-MS ProteinChip array technique in lower-limb deep venous thrombosis, lower extremity varicose veins and normal control, screen biomarker candidates, build diagnostic models and evaluate its clinical significance in early diagnosis of thrombotic diseases. Methods: SELDI-TOF-MS ProteinChip was used to detect the plasma proteomic patterns of 46 cases with lower-limb deep venous thrombosis, 48cases with lower extremity varicose veins and 42 controls. The diagnostic models were developed by bioinformatics tool (nonlinear support vector machine, SVM), and the method of leave-one-out cross validation was used to estimate the discriminating power of patterns. Results : The modelâ… composed by 6 protein peaks could do the best in the diagnosis between lower-limb deep venous thrombosis and lower extremity varicose veins. Its ?sensitivity and specificity were 100%and 97.9%,respectively. The modelâ…¡composed by 3 protein peaks could do the best in the diagnosis between lower-limb deep venous thrombosis and normal control. Its sensitivity and specificity were 95.7%and 95%, respectively. Conclusion: SELDI-TOF-MS is a considerable sensitive and specific protein analytical method which may offer new approach to early diagnosis and differential diagnosis of deep venous thrombosis...
