| ObjectivesTo investigate the effects of berberine on 11β-hydroxysteroid dehydrogenase type 1 expression in the liver tissue in diabetes mellitus rats.MethodsForty SD rats were randomly divided into two groups :⑴Normal Control group(NC,n=10),fed with a standard feed.⑵Diatetes mellitus group(DM,n=30).after fed with high-fat diet for 6 weeks,the rats were injected with streptozotocin (STZ,35mg/kg,ip).And the rats which fasting glucose over 7.0 mmol/l were considered as diabetic rats and randomly divided into two groups again:⑴b erberine-treated DM rats (DM+BBR,n=9).⑵physiological saline-treated DM rats (DM+NS,n=7).The concentration of plasma glucose ,total cholesterol and triglyceride were measured with colorimetry by Automatic Chemistry Analyzer. The mRNA levels of 11β-HSD1 in liver were determined by Reverse Transcription Polymerase Chain Reaction(RT-PCR).ResultsThe fasting plasma glucose ,total cholesterol and triglyceride were higher in the DM group than in the control group(p<0.05).Compared with anterior-posteior the concentration of fasting plasma glucose , total cholesterol and triglyceride were reduced after treated in DM+BBR group(p<0.05),but not in the DM+NS group(p>0.05).The mRNA levels of 11β-HSD1 was higher both in DM+NS group and DM+BBR group than in control group(p<.05).Compared to DM+NS group ,the mRNA expressions of 11β-HSD1 was not prominent in DM+BBR group(p>0.05).ConclusionsBerberine may reduce the concentation of fasting plasma glucose ,cholesterol and triglyceride in type 2 diabetic rats,but it can not affect the 11β-HSD1 mRNA in the liver of type 2 diabetic rats.It suggested that the effects of berberine on blood glucose and lipid is not to achieve by reduced expression of 11β-HSD1 mRNA.
