ObjectiveTo study the best cell ratio of the co-culture system of the rabbit's MSCs and NPCs.MethodsMSCs and NPCs co-culture cells were divided into seven groups by the ratio of 10:1, 5:1, 3:1, 1:1, 1:3, 1:5 and 1:10. Growth condition and morphologic feature of the co-culture cells were observed for seven days, and the cells proliferation was determined by the method of MTT. The synthesis of aggrecan was observed and compared by the method of toluidine blue staining. The expression of typeâ…¡collagen was observed and compared by the method of immunohistochemical staining.ResultsThe co-culture cells in the group of the higher MSCs rate grew faster than others. MSCs and NPCs co-cultured by the ratio of 1:1 and 1:3 were in more active proliferation conditions, and the expressions of aggrecan and typeâ…¡collagen were higher than other group. Differentiations of MSCs to NPCs were observed in the co-culture cells by the ratio of 1:1 and 1:3.ConclusionThe best cell ratio of the co-culture system of the rabbit's MSCs and NPCs is 1:1 or 1:3. The co-culture system induced the differentiation of mesenchymal stem cells.
