The purpose of this study was to prepare cilnidipine liposome and determine itsfeasibility. By using single-factor test and orthogonal test, more appropriate preparationtechnology was established based on the micro examination and the statistics relating to thephysical stability. In accelerated tests, it was found that the liposome with good morphologyand physical stability was produced.HPLC method was adopted to determine both the total content and the entrapmentefficiency of liposome.Various processes concerning the effect of freeze-drying were also investigated.Screening from the different kinds and use levels of cryoprotectant were ensure to protectliposome in freeze-drying. The size distribution and the entrapment efficiency of liposomebefore and after freeze-drying were determined; indicating the liposome after freeze-dryingmainly could maintain their former properties. With appearance, morphology,drug contentand encapsulation efficiency as indicators,the stability of liposome in stress experiment andaccelerated experiment was investigated. All the results suggested that the freeze-driedliposome could mainly enhance the stability in low temperature, and the liposome had betterstability in dark place at low temperature.
