Objects:This study aims to clarify the therapeutic effects of TMP bleomycin-induced pulmonary fibrosis in rats.Methods:40 SD male rats.Randomly divided into sham operation group, model group, positive control group and TMP group, a total of 4 groups of 10 each.Two days post-model,the sham group and control group were treated daily by oral gavage with physiological saline,and the other two groups were fed with Prednisone (1.5mg/kg/d) or TMP(20mg/kg/d).28 days after anesthesia in lung tissues were harvested for follows measured: Body Mass Index;lung quotiety;HE; immunohistochemical staining MMP-2 MMP-9 and western blot MMP-9.Result:1 Results with HE staining under light microscope:Bronchus and alveolus of sham operation group of 28d were normal.Inflammatory of every group reduced distinctly on 28d.A few inflammatory cells remained in part of area.Fibrosis of model group in alveolus interval and around bronchia was more obviously.Fiber appeared coil shape and glomerulus shape.The condition of positive control group was better.Fibrosis degree was not too high in alveolus interval.Fibrosis focus still appeared around bronchus.Inflammatory cells scattered in some alveolus and bronchiole.Pleura thickened.TMP groups became more distinctly.Fiber hyperplastic focus was less in alveolus interval and there weren't inflammatory cells in alveolus.Fibrosis degree was obviously lower than that of model group and positive control group.The results showed TMP had clear curative effect to IPF by reducing fiber piling and restraining forming of fibrosis.2 Immunohistochemical staining:MMP-2 in the normal group was weakly positive; in the model group bronchial epithelial cells and some alveolar macrophages separated strong expression,â…¡-type cells expression. TMP group only in some alveolar septa macrophages positive or weakly positive.Positive expression of the drug group may TMP group. MMP-9 normal lung tissue only in typeâ… alveolar epithelial cells and macrophages with weak expression. Model shows that the lung tissue of bronchial epithelial cells,â… alveolar epithelial cells, fibroblast cells, macrophages and inflammatory cells were strongly positive. Positive control group lung tissue MMP-9 expression was significantly reduced, mainly expressed in bronchial epithelial cells, macrophages and inflammatory cells. TMP group only in bronchial epithelial cells, macrophages,â… alveolar epithelial cells were weakly positive.3 western blot results: Compared with normal group, model group, the lung tissue of MMP-9 levels were significantly increased (P <0.01), compared with model group, TMP group and the positive control group may reduce the pulmonary fibrosis in rat lung tissue MMP-9 levels, and reduce the role of TMP group is more evident.Conclusion:1 TMP can inhibit the expression of MMP-2,MMP-9in early phase in middle and late phases in the cells.2 The western blot of MMP-9 indicate that TMP has the function of anti-fibrosis of lung through restraining the expression of MMP-9.
