The Selective Inhibition Of The LY294002 On The Proliferation In Glioma Cells

Objective Pi3k is the key signaling molecule in life activities, in many tumors it is abnormal and considered the primary pathway of the cell survival. Not only can it promote the tumor cell proliferation ,but also inhibit apoptosis of tumor cells.In this study,w e find the effection and mechanism of the selective interruption on the generation and apoptosis neurogliocytoma cell by treating with LY294002.Methods Targeting the LY294002 to the glioma cell U251. Application of MTT determines glioma cell proliferation; TUNEL determines apoptosis; immunocytochemistry detects the expression of glioma cell proliferation associated protein P27KIP1 and apoptosis protein Caspase9Results The survial rate of the glioma cell with 10umol/L was 0.84±0.02,0.82±0.02,0.57±0.03 by Treating with LY294002 after 4h,8h and 12h,respectively the 30umol/L was 0.73±0.03,0.64±0.04,0.42±0.02,and the 50umol/L was 0.62±0.04,0.42±0.02,0.22±0.01; The apoptotic index at 10umol/L was 5.23±0.21,13.23±0.31,18.23±0.25 by treating with LY294002 after 4h﹑8h and 12h,tat 30umol/L it was 6.40±0.17,15.33±0.42,20.07±0.31,at 50umol/L it was 8.40±0.10,25.17±0.21,32.17±0.21;Expreession of P27KIPIwas positive in plasma of SGS7901 cells and it was positive in plasma and nuclei of cells treated with LY294002;Expreession of Caspase 9 was negative in plasma of SGS7901 cells and it was positive in plasma of cells treated with LY294002. The expression of P27KIP1 in cytoplasm of glioma cells is weak or even not, but the expression is positive in plasma and nuclei of cells by treating with LY294002. The expression of Caspase 9 in cytoplasm of glioma cells is weak or even not, but the expression is positive by treating with LY294002. Conclusions LY294002 not only can significantly inhibit generation of U251 cells,but also can promote apoptosis the mechanisms of U251 cells .The mechanisms may be the positive expression of P27KIP1 and Caspase 9. The positive expression of P27KIP1 can transfer it to the nucleus, and The positive expression of Caspase 9 can generate arrest in G1 phase of cell generation.
