Obejective: To observe the effect of aerobic exercise and standard fodder which has affected the lipid metabolism of the insulin resistance rats which was fed by the fatty and high sugar .Methods:To select 215 clean and healthy male Sprague-Dawley(SD)(purchased from B&K Universal Group Limited )which were 7 weeks age and weighted from 206g to 257g,after adaptive training for a week,the paper divided them into 2 groups stochastically:A,Control,C,n=15,B,Model of Insulin Resistance,MIR,n=200.After buliding the model of IR rats, then divided into 4 groups of MIR,were high fat diet control group(DIO),high fat diet exercise group(DE),ordinary diet group(NOR) and ordinary diet exercise group(NE). Exercise 8 weeks.Measure the blood glucose immediately and the insulin by radioimmunoassay,calculate insulin sensitivity index to determine the model of the insulin resistance in the rat established.After the high fat diet exercise group and ordinary diet exercise group finished its last exercises training,in the next morning the each groups rats were given a general anesthesia with chloralhydrate injection into the abdominal cavity,then were killed. Take the heart of the rat to the fridge of -80°c . Analyze the PPAR-γin cardiac muscle by western blot quantificationally, analyze the morphological changes of the hepar and myocardial cells qualitatively,analyze the adipose change of the hepar and myocardial cells by Oil Red O staining.Result:1.Compared with the control group,The insulin sensitivity index of MIR group was lower significantly. Through aerobic exercise and / or to change normal diet,insulin sensitivity index was increased significantly.High fat diet can induce insulin resistance in the rats.2.The fat cells of liver is dyed a deep red by Oil Red O staining. The fat cells of the liver of the rats of the control group spread out evenly, however, the fat cells of the liver of the rats of the high fat diet group are significantly increased, with masses to red lipid drops.After aerobic exercise and/or dietary intervention,Compared with the high fat diet group ,the fat cells of the liver of the three intervention groups decreased significantly .Compared with the control group,the fat cells of the liver of the ordinary diet group and the ordinary diet exercise group are close to the normal ones.Compared with the high fat diet group,the high fat diet exercise group decreased significantly,too.3.The changes in the PPAR-γof cardiac muscle after intervention:Compared with the control group,the expression of the PPAR-γof the high fat diet exercise group and the ordinary diet exercise group increased significantly(P<0.05),but the expression of the PPAR-γof the high fat diet group decreased significantly(P<0.05);Compared with the high fat diet group ,the expression of the PPAR-γof the high fat diet exercise group and the ordinary diet exercise group increased significantly(P<0.05),although the expression of the PPAR-γof the ordinary diet group increased,however,there is no statistical significance.Conclusions :1.After high fat diet of 8 weeks to SD rats, we can establish animal models of insulin resistance. 2.Aerobic exercise and the change to regular diet both can improve the condition of IR significantly.
