Research background Insulin resistance is a common pathological condition, has close relationship with many diseases.Among them, patients with hypertension easily combined with insulin resistance. In previous studies, most of the studies focused on the study of vascular insulin resistance and spontaneous hypertension.However, recent studies have indicated that there is a certain relationship between myocardial insulin resistance and the expression level of myocardial protein in hypertensive rats.Clinical studies have confirmed that aerobic exercise is a very safe and effective to improve insulin.But on aerobic exercise in the study is relatively less in spontaneously hypertensive animal myocardial insulin resistance effect, the effect and the specific mechanism of action, but also the lack of systematic analysis.Objective 1.To observe the effect of aerobic exercise on blood pressure in hypertensive rats. 2.To observe the effect of aerobic exercise on insulin sensitivity in hypertensive rats.3.To observe the effect of aerobic exercise on myocardial protein in hypertensive rats.Method Spontaneously hypertensive rats(SHR) and Wistar-Kyoto rats(WKY) were used as experimental animals, and the long-term aerobic exercise model was induced in rats,Blood pressure levels and myocardial protein expression levels were measured in rats, and the effect of aerobic exercise on insulin sensitivity was analyzed.Results At different time, the weight of the three groups were detected, and the weight of the three groups were all increased.At the beginning of the 4 week, the body weight of the SHR swimming training group was significantly lower than that of the SHR motion control group, both P < 0.05.SHR motion control group and SHR swimming training group in systolic and diastolic blood pressure were significantly higher than that of WKY group at different time, both P < 0.05.The systolic pressure and diastolic blood pressure of SHR swimming training group were significantly lower than the average level of SHR motion control group at different time, P < 0.05.Glucose tolerance test results showed that the blood glucose of three groups rats reached the maximum for 30 min,then the blood glucose began to peak off,at 60 min, 90 min, 120 min,the blood glucose of SHR swimming training group was significantly lower than the average level of motion control group, P <0.05.Compared to WKY group,left ventricular end diastolic pressure(LVEDP) and resting heart rate of SHR swimming training group were significantly decreased, left ventricular systolic pressure(LVSP) increased significantly, P < 0.05.But the differentiation of left ventricular systolic / diastolic pressure did not change significantly, compared with the WKY group, P > 0.05.Then the three groups rats were treated with insulin for 30 min, compared with SHR motion control group, WKY and SHR swimming training group, the differentiations of left ventricular systolic/diastolic pressure were significantly increased,P < 0.05.The expression levels of Akt and PPAR? in the SHR motion control group were significantly lower than those in the SHR swimming training group and the WKY group, both P < 0.05.Conclusion In spontaneously hypertensive rats, there are some phenomena of myocardial insulin resistance.The effect of aerobic exercise on hypertensive myocardial insulin sensitivity can be effectively improved in spontaneously hypertensive rats.Analysis of the relevant mechanisms, it may be affected by the decline in the expression of PPAR? and the damage of PI3K-Akt signal.
