Objectives: Our aims of this study are to analysis the levels of the 5-HT and explore expression levels of 5-HT3B receptor in different brain regions;seek for parts of the mechanisms of angry in;and study the effect of Jingqianshu granule acting on anger- in.Methods:We made the model rats with anger in by the methods of resident-intruder paradigm and social isolation,estimated model rats by open-field test and sucrose preference-test,and some of them were treated with Jingqianshu granule . We divided into anger in and anger out model by median statistics after doing the experiment of the resident-intruder for five days, and the anger-in model rats were divided into model group and administration group .The administration group rats were treated with Jingqianshu granule .After two weeks,we killed the rats by decapitation and dissected Frontal cortex,hippocampus,hypothalamus and Parietal cortex of the brains.We detected the expression levels of 5-HT3B mRNA in different brain regions of rats in groups including the normal group,the model group(resident intruder paradigm model group) and administration group by using a semi-quantitative RT-PCR technology. After total proteins were extracted from these brain tissues,protein levels of 5-HT3B receptor were detected in the encephalic regions with western blot method.The 5-HT levels in different brain regions were detected by the method of high pressure liquid chromatogram (HPLC).Results: After a week,compared with the blank group, the score of model group including the crossing score, rearing score and total score were markedly increase(P<0.01);saccharine preference index reduced(P<0.01). When treating with Jingqianshu graulne,the score of open-field markedly decrease(P<0.05) and saccharine preference index markedly increased(P<0.05).RT-PCR results: The mRNA expression of 5-HTR3B in different brain regions of model rats increased obviously (P<0.01),comparing with the normal control group. When treating with Jingqianshu graulne, the mRNA expression of 5-HTR3B decrease obviously (P<0.01)Western-blot result: The expression of 5-HTR3B in different brain regions of model rats increased obviously (P<0.01),comparing with the normal control group.. When treating with Jingqianshu graulne, the expression of 5-HTR3B decrease obviously (P<0.01)HPLC-FLU result: The contents of 5-HT in different brain regions of model rats decreased obviously(P<0.01), comparing with the normal control group. When treating with Jingqianshu graulne, the contents of 5-HT increase obviously (P<0.01).Conclusion: We found the method by resident intruder paradigm could simulate the model of Worried and angry effectively. The expressions changes of the 5-HTR3B and the content of the 5-HT in different brain are in close relation to Worried and angry Rats. At the same time,the Jingqianshu granule Call down-regulate the 5-HT3B expression levels and 5-HT content of model rats in different brain regions,which maybe one of the mechanism to treat Worried and angry.
