Objective:Our aim is to investigate the relationship between the expression levels of 5-HT1A receptor mRNA and protein in different brain regions(hypothalamus and hippocampus)and the pathogenesis of PMS with liver-qi depression,as well as to study the rejustion effects of Jingqianshu granule on the syndrome.Methods:Confirmed the estrum of rats by observing the vagina cell shape with 40 times zoom microscope and then selected the rats in non-acceptance period to perform model of PMS with liver-qi depression by bounding its feet. The open-field experiment was used to evaluate the model. We detected the expression levels of 5-HT1A receptor mRNA and protein in different brain regions of rats in groups including the control group, the model group (bound model group)and treatment group by using a semi-quantitative RT-PCR gene amplification technology and western blot.Results:The results of open-field experiment showed that there was significant difference (P<0.05) espectively between the model group and the control group as well as the treatment group. However, there was no significant difference between the control group and the trearment group in open-field experiment.There was significant difference (P <0.05) between the model group and the control group,the expression levels of 5-HT1A receptor mRNA and protein in hypothalamus and hippocampus of model rats were obviously lower compared with the control group; There was significant difference (P< 0.05) between the model group and the Jingqianshu granule group,the expression levels of 5-HT1A receptor mRNA and protein of the Jingqianshu granule group were statistically higher than model group, and had no difference (P>0.05)compared with the control group.Conclusion:The modle rats effectively simulate the clinical abnormalities of PMS with liver-qi depression. Lower expressions level of the 5-HT1A receptor mRNA and protein are in close relation to PMS with liver-qi depression. At the same time, the Jingqianshu granule can up-regulate the 5-HT1A receptor mRNA and protein expression levels of model rats in different brain regions, which may be one of the mechanisms to treat PMS with liver-qi depression.
