This study was to find the right compounds as an indicator of Cucumis melo quality control. the ethanol extract of Cucumis melo isolated four compounds, through chemical and spectral analysis, they were identified cucurbitacin A, cucurbitacin B, cucurbitacin B 2 O glucoside and cucurbitacin E, Among them, cucurbitacin A was isolated from Cucumis melo for the first time.cucurbitacin B extraction process was studied as follows: ethanol extraction method: 11 times the amount of 65% ethanol extract 3 times, each half an hour. The process proceeds under the cucurbitacin B relative extraction rate of 86.15%. The method is feasible and reliable.Because many different types of cucurbitacin and their chemical structure very similar, led to great difficulties in the separation and purification process . This is the first use of the structural transformation, through hydrolysis, microbial transformation, acid hydrolysis of three methods of investigation, in which enzyme used IM, PS-D, L-1754, N435, F-AM, and lipase which 6 enzyme to cucurbitacin B-2-O glucoside were hydrolyzed showed the content of cucurbitacin B has not changed. Microbial transformation with the E. coli, yeast and molds to cucurbitacin B-2-O-glucoside were transformed, the results show that cucurbitacin B content is not changed. Final choice of the acid hydrolysis method, a low concentration of acetic acid and hydrochloric acid to its hydrolysis, the content of cucurbitacin B has increased. Therefore, the method preferred by acid hydrolysis. By acid hydrolysis of cucurbitacin B-2-glucoside into cucurbitacin B, cucurbitacin B was increased to 7%. In addition, the method by alkaline hydrolysis to extract from Astragalus membranaceus(Fisch), Astragaloside extracted than the traditional method of extracting Astragaloside, the extraction rate has been increased significantly. Further evidence of structural transformation in the feasibility of extraction of natural products.Macroporous resin with cucurbitacin B were isolated and purified of the AB-8, XAD-16, D101, HPD-600 resin cucurbitacin B of four adsorption rate and desorption rate, optimized XAD-16 resin cucurbitacin B were isolated to determine the adsorption and elution conditions, the results showed that after XAD-16 resin concentration, the higher the recovery rate of cucurbitacin B, and the purity was improved, effectively remove the impurities, experiments to intended purpose. cucurbitacin A, cucurbitacin B, cucurbitacin B-2-O glucoside and cucurbitacin E were further tested for cytotoxicity against two human cancer cell lines(Hela and HepG-2), results showed that: cucurbitacin A and cucurbitacin B on Hela cells and HepG-2 cells better inhibition.
