| Background In recent years, along with diet and lifestyle changes, type 2 diabetes (T2DM) in young people is in high morbidity. In the antient Chinese medical records , the records of diabetes mellitus therapy is too rich to be found to cure T2DM ,so that it is valueble and practical for us to explore it.Objective To Investigate, of diabetic rats, the effects of Astragalus compound particles comes from Prof. La Wanying on Glucose and lipid metabolism , the pancreas and morphological changes, the pancreas immunolistochemical changes of Insulin-like growth factor(IGF).Methods To establish T2DM animal model ,intraperitoneal injected streptoptocin(STZ) 40mg/kg to SPF-class male wistar rats.after randomization, different groups established:the normal control group, diabetic control group, medicine comparison group (repaglinide),traditional Chinese medicine comparion group(shen qi jiangtang keli ) and Astragalus compound particles group (ACP,ACP01: High-dose ACP group, ACP02: Medium dose ACP group and ACP03: low-dose ACP group). With different administration for 4 weeks, body weight were measured at 2nd week and 4th week. at the end of animal expriments, the rats were killed while the collection of blood for fasting blood glucose(FBG), Triglyceride(TG), total cholesterol (CHO),high-density lipoprotein cholesterol(HDL-C) and low density lipoprotein cholesterol(LDL-C) and collection of blood for fasting insulins(FINS) with radioimmunoassay and fixation of pacreasis for morphlogical and immunohistochemical tests for IGF. Results 1.rats nutritional status: compared with the normal control group, diabetic control group, medicine comparison group, Traditional Chinese medicine comparison group and ACP01~03 group rats had signicant manifestations such as polydipsia,polyuria,fur wither and weight loss or retardation;and the body mass index of these groups significantly decreased compared with normal control group(p<0.05)2. biochemical and hemorheology tests: (1)compared with the normal control group, diabetic control group and ACP(01~03) groups rats had significantly higher blood glucose, LDL-C,TG,CHO,Glycated hemoglobin( HbA1c) levels and lower HDL-C level as well as higher Whole Blood High Shear Rate , Low Shear Rate(p<0.05,p<0.01). compared with the normal control group, medicine group and traditional Chinede meidicine group rats had significantly differences such as higher blood glucose, LDL-C,TG,CHO,Glycated hemoglobin( HbA1c) levels(p<0.05,p<0.01) while had not difference with higher Whole Blood High Shear Rate , Low Shear Rate(p>0.05). compared with other interwention groups, ACP groups had lower blood glucose(p<0.01),ACP01 groups had the lower blood glucose and HbA1c level compared with other ACP groups(p<0.01).(2)FINS and insulin action index(IAI): compared with the normal control group, other groups rats had significantly higher FINS and IAI negative increase(p<0.01). compared with the diabetic control group, medicine group and traditional Chinede meidicine group rats had not difference with FINS and IAI(p>0.05),but difference of FINS and IAI were observed btween ACP groups and diabetic control group(p<0.01).And no difference of FINS among ACP groups(p>0.05)while ACP01 group had reduced IAI negative compared with ACP01 and ACP02 group(p<0.01). 3.morphlogical changes:(1)HE staining:compared with normal control group, in other groups rats pancreatic tissues, different degrees of hyaline degeneration,fatty degeneration and inflammatory cell infiltration were observed, ,but those pathological changes in diabetic group were more serious significantly than other intervention groups rats.(2)immunohistochemical staining: compared with normal control group, in other groups rats pancreatic tissues, different degrees of positive expression of IGF . but those changes in diabetic group were more serious significantly than other intervention groups rats(p<0.05,p<0.01).Conclusion Astragalus compound particle(sACP) have rugulating effects on glucose and lipid metobolism of Qi and Yin Deficiency diabetic rats that maybe achieved through improve pancreatic lesion ,improve insulin resistance and IGF expression. |
