Objective:To observe different doses of APS on the Insulin resistance in type 2 diabetes rat modle which caused by High glucose and lipid combined Low-dose streptozotocin and the influence of serum TNF-a,1 eptin and adiponectin levels,To explore the APS on the role of the symptoms and insulin resistance of type 2 diabetes rats and the mechanism about it.Methods:To build type 2 diabetic rat model of insulin resistance which feeding SD rats with High sugar high fat diet for 4 weeks after the low dose first injection of streptozotocin.The SD rats were randomly divided into normal control group, diabetic+saline control group,diabetes+ high-dose APS treatment group,diabetes+medium-dose APS treatment group,diabetes+ low-dose APS treatment group.Examined fasting blood glucose (FPG), fasting plasma insulin(FINS), triglyceride (TG), high density lipoprotein (HDL), low density lipoprotein (LDL), cholesterol (TC) and insulin sensitivity index (ISI), insulin secretion index (IS), insulin resistance index (IRI) and serum TNF-a, Leptin and adiponectin levels.Results:1,The method about high glucose and lipid feeding with low dose streptozotocin injection successfully established type 2 diabetic rat model of insulin resistance.2,Compared with the diabetic saline control group,blood glucose levels were significantly lowerin APS medium and high-dose treatment group.Blood insulin levels in APS high, medium and low dose treatment group also decreased significantly which compared with the diabetic control group.3,Compared with the diabetic saline control group, serum TG, TC, LDL levels decreased significantly in APS high, medium and low dose treatment group,,while serum HDL was significantly increased.4,Compared with the diabetic saline control group, APS high, medium and low dose treatment group rats increased insulin sensitivity index, insulin resistance index decreased, indicating that the symptoms of insulin resistance.5,Compared with the diabetic saline control group, the serum TNF-a and Leptin levels were significantly lower in APS high, medium and low dose treatment group, while adiponectin levels were significantly increased.Conclusion:1.APS may play a very good therapeutic effect on type 2 diabetic rats,manifested as reduced blood glucose, improved glucose tolerance, improved peripheral tissue sensitivity to insulin and reduced insulin resistance.2.APS can significantly improved the lipid abnormalities which type 2 diabetic ratsassociated with.3. APS on the treatment of diabetes and insulin resistance is related to inhibiting the production of inflammatory cytokines TNF-a, leptin, increasing adiponectin production, improving insulin signal transduction pathway.
