Objectives To investigate the effects of Additional Xiao Xianxiong decoction on myeloperoxidase (MPO),tumor necrosis factor-α(TNF-α) and C-reactive protein(CRP) during myocardial ischemia/reperfusion injury in rabbit and its protective mechanism.and then provide evidence for treatment of myocardial infarction etc in clinic.Methods 1 animal; healthy male Japanese rabbits(weight 2.5-3.0Kg) ,with big ears,feed in barrier circumstance,eat freedom.Which provide in North china coal medical university aniamal experiment center【licencenumber scxk(京)2006-0005】.2 Divide group; 32 healthy male Japanese rabbits(weight 2.5-3.0Kg) ,with big ears were randomly divided into four groups,sham operated group(group A ),myocardial ischemia group(group B),Xiao Xian Xiong Tang adding tastes low dosage group(group C), Xiao Xian Xiong Tang add taste high dosage group(group D).3 Give medicine;Each rabbit of group D was intragastricly administrated Xiao Xian Xiong Tang adding tastes,according to eight times of person's dosage/Kg/day(6.32g/Kg),as the same method of group D, the dosage of group C was 3.16g/Kg,The group A and B was intragastricly administrated physiological saline。Each group was intragastricly administrated for five days before building model.1 time/day.4 build model; (1)Group A,the rabbit's left anterior descending coronary artery(LAD) was false ligated,(2)group B,C and D ,the LAD was ligated for 30 minutes, then opened it.The reperfusion were finished in 120mins.5 measure; MPO in plasma was measured by radio-immunity structure histochemistry and biochemical method, TNF-αin plasma was detected by radio-immunity method, CRP in plasma was measured by radio-immunity compare with chaotic method. Result the concentration of MPO in group B,C,D markedly increased than group A(P<0.01),the concentration in group C decreased than group B( P<0.05 ) ,and in group D significantly decreased than groupB(P<0.01),although it in group D slightly decreased than it in group C,the level was no significant difference(P>0.05).the concentration of TNF-αin group B,C,D markedly increased than group A(P<0.01), the concentration in both group C and D decreased than group B(P <0.05), although it in group D slightly decreased than it in group C ,the level was no significant difference(P>0.05).the concentration of CRP in group B,C,D markedly increased than group A(P <0.01), the concentration in both group C and D decreased than group B(P <0.05), although it in group D slightly decreased than group C,the change was no significant difference(P >0.05).Observaions of pathological;Group A myocardial fibre arranged in order,borderline was intergitey. Group B lots of myocardial cells edemaed,vacuoled changing and partly endochylema catched lightly,the shape of cardiac muscles were wavilness, inflammatory cell infiltrated significantly. Group C myocardial cells edemaed lightly, endochylema distributed nonuniformly, inflammatory cell infiltrated lightly. Group D most myocardial cells edemaed lightly, endochylema distributed nonuniformly, some cells had vacuoled changes,inflammatory cell infiltrated lightly.Conclusion Xiao Xian Xiong Tang adding tastes may protect the cardiac muscles injuried by ischemia and reperfusion myocardial effectively,which might be associated with decreasing the level of inflammation MPOandCRP released ,which might be associated with alleviating the impairment of oxygen free radicals and TNF-α.
