Objective: To investigate the effect of capecitabine to human esophageal epithelium cell line SHEE and its cancerization cell line SHEEC, and the relationship with thymidine phosphorylase (TP); providing experimental data and evidence for capecitabine applicating in chemotherapy of esophageal carcinoma.Methods: The expressions of TP in SHEE,SHEEC were detected by immunohistochemistry method, then compared with which in esophageal carcinoma tissue and normal esophageal epithelial tissue beside the carcinoma. Detecting the concentration of metabolins (Fluorouracil and Doxifluridine) in human blood serum collected after oral application of capecitabine, preparing blood serum for cell experiment. MTT assay was performed to evaluate the influence of human blood serum collected after oral application of capecitabine, Fluorouracil and Doxifluridine on cell multiplication of SHEE and SHEEC cells.Results: TP was negative in SHEE cell line, positive in SHEEC cell lines. The positive staining was mainly observed in the cytoplasm regions, while weak staining was also observed in the nuclear of some cells. TP was negative in normal esophageal epithelial tissue; in esophageal carcinoma tissue, the positive expression rate of TP was 76.09% (35/46). The positive staining was mainly observed in the cytoplasm regions, and also observed in the intercellular substance. The concentration of Fluorouracil and Doxifluridine in human blood serum collected after oral application of capecitabine(blood serum contained drugs) were 103.75ng/ml,2.278ug/ml; there was no Fluorouracil and Doxifluridine in human blood serum collected before oral application of capecitabine. In certain range, blood serum contained drugs, Fluorouracil and Doxifluridine to SHEEC, blood serum contained drugs, Fluorouracil to SHEE, inhibited the cell growth in a concentration and time-dependent manner, growth inhibiting ratio achieved the peak at 48h; Doxifluridine had no obvious growth inhibition effect to SHEE cell line; the growth inhibiting ratios of SHEE and SHEEC cells using with 10% blood serum contained drugs were 11.23% and 23.63% at 48h, it was significantly different (P<0.05); The growth inhibiting ratios of SHEE and SHEEC cells using with 10.375ng/ml Fluorouracil, which concentration was same as Fluorouracil in the group of 10% blood serum contained drugs, were 10.57% and 9.98% at 48h, it was not significantly different (P>0.05); the growth inhibiting ratios of SHEE and SHEEC cells using with 227.8ng/ml Doxifluridine, which concentration was same as Doxifluridine in the group of 10% blood serum contained drugs, were 1.69% and 16.76% at 48h, it was significantly different (P<0.05); the growth inhibiting ratios of SHEE and SHEEC cells using with 10.375ng/ml Fluorouracil plus 227.8ng/ml Doxifluridine, which concentration was same as Fluorouracil plus Doxifluridine in the group of 10% blood serum contained drugs, were 1.69% and 16.76% at 48h, it was significantly different (P<0.05).Conclusion: Cancerization of normal esophageal epithelium cell follows with increase of positive expression of TP. Capatibine has weak inhibition effect to SHEE cell line, selective lethal effect to SHEEC cell line. Expression of TP have no obvious correlation with chemo effect of 5-Fu.
