Objective: To investigate the effects of Pioglitazone Hydrochloride on gene expression of ATP binding cassette transporter A1, ATP binding cassette transporter G1 and scavenger receptor class B type I in THP-1 derived Macrophages.Methods: 1.THP-1 cells were transformed into Macrophages in vitro. 2. After exposure of the cultured THP-1 derived macrophages to Pioglitazone Hydrochloride at different concentration(0uM,1uM,10uM,20uM,30uM) for 24 h ABCA1, ABCG1and SR-BI mRNA expression were observed by Realtime PCR. 3. THP-1 derived macrophages were incubated with Pioglitazone Hydrochloride (10uM), or preincubated with GW9662(2uM) for 1 hour before stimulation with Pioglitazone Hydrochloride. ABCA1, ABCG1and SR-BI mRNA expression were analyzed by Realtime PCR after 12h and 24 h of treatment, and their protein expression were detected by Western Blot after 24h and 48 h of treatment.Results: 1. Pioglitazone Hydrochloride significantly increased ABCA1, ABCG1and SR-BI mRNA expression after 12h and 24h of treatment in THP-1 derived macrophages(P<0.01) as well as increased their protein expression after 24h and 48h of treatment(P<0.01). 2. After stimulation by Pioglitazone Hydrochloride with different concentration for 24h ABCA1, ABCG1and SR-BI mRNA expression of macrophage were markedly increased which showed a does-dependent manner (all P<0.01). 3. GW9662 obviously depressed the raise of ABCA1, ABCG1and SR-BI mRNA and protein expression in macrophages which were induced by Pioglitazone Hydrochloride (P<0.01).Conclusion: Pioglitazone Hydrochloride increased the level of ABCA1, ABCG1and SR-BI mRNA and protein expression in THP-1 derived macrophages by activating PPARγ.
