Objective Our object is to understand whether there is a difference of 5-HT4 receptor expression in proximal and distal colon of Hirschsprung disease and Hirschsprung's allied disorder, and normal colon were compared. The relationship between to understand 5-HT4 receptor and HD /HAD.Methods We collected 15 cases of Hirschsprung's disease specimens, 10 cases of Hirschsprung's allied disorder specimens and 10 normal colon specimens.Use SP immunohistochemical staining to understand receptor sites and different levels of expression. Results In Hirschsprung's disease receptors in the mucosa in the distal end decreased compared with the proximal end while receptors were not expressed in the myenteric plexus of the proximal end,.In Hirschsprung's allied disease the number of receptors also decreased in the distal mucosa and myenteric plexus. Compared with the normal colon receptor expression decreased in HD/HAD.Conclusion We confirmed 5-HT4 receptor expression was different in proximal and distal end in HD/HAD.Receptors were expressed higher in the proximal end, but were lower than the normal control group. HD distal section without ganglion cells between the muscle had no expression.5-HT4 receptors in Hirschsprung's disease and Hirschsprung's allied disease may play a role in etiology or play one of factors aggravating the symptoms. Detection of 5-HT4 receptors in HD/HAD can be used as reference for preoperative differential diagnosis and provide a new direct for clinical treatment.
