Study On The Role And Mechanism Of P55PIK In Tumor Invasion And Metastasis

Objective Our object is to detect the different expression level of p55PIK in the Colorectal Cancer and normal epithelium tissue, then investigate the relationship of the p55PIK with the development of Colorectal Cancer and the cell migratory ability.Methods The 55 samples of Colorectal Cancer and normal epithelium tissue are collected, and detect the expression level of p55PIK by Real-time PCR and immunohistochemistry (IHC). Then construct the expression vector pFlag-CMV4- p55PIK, the migratory ability are observed in Colorectal Cancer cells after over expressing p55PIK.Results The Real-time PCR results shows that the transcription level of p55PIK is up-regulated in carcinoma (63.6%), while down-regulated portion is 16.4%. The consequences of immunohistochemistry coincide with the Real-time PCR. The expression vector of pFlag-CMV4- p55PIK is constructed successfully, both the cell migratory ability are elevated conspicuously after over expressing p55PIK.Conclusion our experiments identified that most of Colorectal Cancer are over expressing of p55PIK, and also the overexpression of p55PIK improve the migration capability of Colorectal Cancer cells.
