Objective: To investigate the expressions of PD-1 in human gastric carcinoma and normal gastric mucous surrounding the tumor, and explore their correlation to clinical pathological features of gastric carcinoma. To investigate the cellular localization of PD-1+ T-lymphocyte, and the capability of proliferation and cytokine production of CD8+ T-lymphocyte in human gastric carcinoma.Methods: The expressions of PD-1 in 68 cases of gastric carcinoma and corresponding normal gastric mucous were detected by SABC immunohistochemistry. The cellular localization of PD-1+ T-cell was detected by fluorescence immunohistochemistry. The capability of proliferation and cytokine production of PD-1+ T-lymphocyte were detected by flow cytometry.Results: 53 of the 68 gastric carcinoma specimens(77.9%) were positive in PD-1 contrast to 9 of the 68 normal gastric mucous (13.2%) were positive in PD-1. The expression of PD-1 was correlated with TNM types, invasion depth and lymph node metastasis (p<0.05) and was not correlated with sexuality, age, area, and location of tumor (p>0.05). Fluorescence immunohistochemistry and flow cytometry confirmed that PD-1 was mainly expressed on the CD8+ T-cell, the capability of proliferation and cytokine production of of PD-1+CD8+ T-cell became lower than the normal CD8+ T-lymphocyte.Conclusion: The expression of PD-1 in gastric carcinoma is more than that in normal gastric mucous surrounding the tumor. The expression of PD-1 in gastric cancer was mainly CD8 + T cells, high expression of PD-1 T-lymphocyte proliferation and cytokine secretion capacity were reduced, so the function of PD-1+ T-lymphocyte is suppressed.
