Plasma Lipidomics Analysis Of HLP Patients With "Gan-yu-pi-xu" Syndrome

Objectives: Seek for the lipid biomarkers of lipidomics for HLP patients with"gan-yu-pi-xu"syndrome by study the lipidomics of HLP with"gan-yu-pi-xu"syndrome. To analysis the foundation of HLP with"gan-yu-pi-xu"syndrome and provide the objective basis of HLP with"gan-yu-pi-xu"syndrome by lipidomics.Methodology: In this study, we start with the clinclal research of the HLP syndromes. On this basis combined with early clinical research and documentation of the findings of our group, we choose one of the common sydrmomes of HLP,"gan-yu-pi-xu"syndrome, as our lipidomics study subject. Devide the HLP group into"gan-yu-pi-xu"group and none"gan-yu-pi-xu"group, and collect the normal person as the control group. According to < Guidelines for prevention of Dyslipidemia in adults in China>(2007), , (2002) and < Specification for clinical diagnosis and treatment of TCM>, set this research's acceptance and elimination standard. We kept basic information of the patients with HLP in the form of questionnaires, and dialectically categorize the accepted patients'TCM syndromes. Test their blood lipids of HLP patients by biochemical analyzer and conduct statistical analysis for pertinence of different TCM syndromes and blood lipids indexes with the help of SPSS18.0. Filter the suitable patients and collect the blood samples. Analyze the lipid metabolism in the blood of HLP patients and normal people by UPLC/Q-Tof MS. Educe UPLC/Q-Tof MS result via MasslynxV4.1. Conduct PCA and PLS-DA by SIMCA-P.Result: 1 Dialectically categorize the TCM syndromes of 82 HLP patients in the clinical research, we find out 17 patients are with"gan-yu-pi-xu"syndrome(20.73%), 13 patients are with"gan-shen-yin-su"syndrome(15.85%), 11 patients are"pi-shen-yang-xu"(13.41%), 8 patients with"tan-zhuo-zu-e"syndrome(9.75%), 8 pateints with"qi-zhi-xue-yu"syndrome(9.75%), 9 pateints with"yin-xu-yang-kang"syndrome(10.98%) and 16 pateints are with other syndrome(19.51%). 2 Compare each syndrome's blood lipid, we find out"qi-zhi-xue-yu"group has higher TC,LDLC level than"pi-shen-yang-xu"and"gan-yu-pi-xu"group(P<0.05), higher ApoB level than"gan-shen-yin-xu"and"pi-shen-yang-xu"group(P<0.05). The"tan-zhuo-zu-e"group has higher LDLC level than"pi-shen-yang-xu"group(P=0.018), other syndromes' comparison of blood lipid has no statistical differences (P >0.05).The"gan-yu-pi-xu"group accepted in the lipidomics research has lower TC,LDLC level compared to none"gan-yu-pi-xu"group(P<0.05). 3 From lipid analysis of normal group,"gan-yu-pi-xu"group and none"gan-yu-pi-xu"group, we can notice obvious difference in regards lipids composition. There are 6 lipid biomarkers of HLP, might be PS, PC, PE, FA and SP. The content of PC, PS, and PE are lower than the normal group. We also find out there are 4 lipid biomarkers for HLP with"gan-yu-pi-xu"syndromes as a result of analysis, might be PG, PS, FA and SP. The content of PS is lower than other syndrome.Conclusion: 1 Hyperlipemia with"gan-yu-pi-xu"syndrome is the main TCM syndromes in clinical research. 2 The number of lipid biomarkers for HLP is six, and the number of lipid markers for HLP"gan-yu-pi-xu"syndrome is four.
