Objective This study is to detecting the expression of ERCC1 and LRP in non-small cell lung cancer and its adjacent tissues by the way of immunohistochemistry.In order to find out the relationship between the expression of ERCC1/LRP in NSCLC and its adjacent tissues,we discuss the relationship between ERCC1/LRP and pathological features of NSCLC combined with clinicopathological factors.The purpose is to provide a theoretical basis for clinical individualized treatment.Methods The two-step immunohistochemical way is used to detect the expression of ERCC1 and LRP in 45 non-small cell lung cancer tissues and 21 adjacent tissues.Using SPSS 17.0 statistical software for data analysis.Theχ2 test or fisher exact test is used to analyze the relationship between the expression of ERCC1/ LRP in NSCLC and clinical pathological features;Spearman correlation test is used to detect the correlation between ERCC1 and LRP.Takeα=0.05 as significant level.Results①The positive expression rate of ERCC1 in 45 patients with non-small cell lung cancer and 21 cases of adjacent tissues is 55.6% (25/45) and 80.9% (17/21),the difference is statistically significant (P <0.05); The positive rates of pathological types are: adenocarcinoma 73.7% (14/19)> squamous cell carcinoma 42.3% (11/26), the difference between groups is statistically significant;The expression levels of ERCC1 are not related with gender,age,tumor differentiation, lymph node metastasis and clinical stage(P> 0.05)②The positive expression rate of LRP in 45 patients with non-small cell lung cancer and 21 cases of adjacent tissues is 66.7 % (30/45)and 38.1%(8/21),the difference between groups is statistically significant;The positive rates of each pathological types are:adenocarcinoma 84.2% (16/19)>squamous cell carcinoma 53.8% (14/26),the difference between groups is statistically significant.The expression level of LRP is related with tumor differentiation.The expression of LRP in well-differentiated group was 77.4%, while in poorly differentiated group was 42.9%,the difference between groups is statistically significant.But there are no significant differences with gender,age,clinical stage and lymph node metastasis (P>0.05).③The expression of ERCC1 and LRP in the 45 cases of non-small cell lung cancer is positively correlated(r=0.316, P=0.034).Conclusions①The expression of ERCC1 in NSCLC adjacent tissues is significantly higher than in NSCLC tissues.But the expression of LRP in NSCLC tissues is significantly higher than adjacent tissues.Reducing the expression level of ERCC1 suggest an increased risk of cancer.②The expression of ERCC1 and LRP in lung adenocarcinoma is higher than in squamous cell carcinoma of the lung.It is suggested that the pathological type must have some correlation with the expression of ERCC1 and LRP.③The expression of LRP is also related with histological gradings.Well-differentiated group is higher than poorly differentiated group.LRP is one of the main explanations of multidrug resistance in NSCLC .It indicate that the expression of ERCC1 and LRP is related with differentiation degree. Comparing to the expression of ERCC1/LRP in NSCLC and its adjacent tissues,concerning the relationship between LRP/ERCC1 and clinical pathological characteristics of the NSCLC,there must be some correlation between drug resistance of non-small cell lung cancer and the expression of ERCC1/ LRP.ERCC1 and LRP is expected to be the potential predicted molecular markers of anticipating the efficacy of chemotherapy.It can provide a theoretical basis for developing the scientific and effective treatment and clinical programs to individualized treatment.
