Objective: To investigate the hypoglycemic effect and the islet cell functional regulation of deuterium-depleted liquor on Rats with diabetes mellitus and islet cells (NIT-1). Methods: In vivo: Wistar rats were induced by streptozotocin (STZ) one-time through intraperitoneal injection to create diabetes models. The experimental rats were divided into seven groups (ten each) according to the means of intervention: the control group(A) (without STZ inducing, normal water, normal water);the model group(B) (intragastric administration of normal saline, fed with normal water); DDW group(C) (DDW, DDW); normal Chinese liquor group(D or E)(one unit (low) or six unit (high) Chinese liquor, normal water); deuterium-depleted Chinese liquor group(F or G)( one unit (low) or six unit (high) Chinese liquor, DDW). The oral administration of liquor solution was 0.01 ml/g (volume/body weight) each day. After four-week intervention, the blood was collected, removed the pancreatic and observed the pathology structure. The fasting blood glucose (FBG) and fasting insulin level were measured. In vitro: The concentration of liquor was 150mmol/L. NIT-1 cells were divided into 7 groups as the same with experiments in vivo.STZ was added at 22mmol/L and kept 30min. The concentration of Chinese liquor was 150mmol/L(low) and 300mmol/L(high) and kept 12h. Then the level of SOD and MDA was measured, meanwhile the pro-apoptotic effect was analyzed by TUNEL staining and Annexin V-FITC staining. Results: In vivo: FBG in each group followed the order: E>B>D>G>C>F>A, group C and group D both have significant difference from other groups (P<0.05). FINS of each group followed the order: A> F > G > D> C> E >B, except E, other groups have significant difference from the model group(P<0.05). The histological examination showed that in group B ,group D and group E, the pancreatic islet cells were decreased and coupled with hydropic degeneration and vacuolar degeneration.DDW had obvious protective effects on some structural abnormalities of pancreatic isles induced by Chinese liquor. In vitro: The results show the same result: The control group was significant difference from other groups (P<0.05); the group F was significant difference from other groups(highest SOD level; lowest MDA level; had the least number of the pro-apoptotic effect (P<0.05) Conclusion: The fact that the deuterium depleted liquor has significant effect of decreasing glucose levels, due to increasing of the plasma insulin concentration, confirmed that the deuterium-depleted water could reduce the damage caused by excessive drinking.
