1. ObjectiveIn nature, the volume fraction of deuterium in water is 0.015% and the water below this level is called deuterium-depleted water(DDW).Different physical and chemical properties depend on different quality between these two nuclides, protium and deuterium. Some studies show that 25%-30% of deuterium water for drinking can increase the survival of ascites tumor, inhibit fibroblast L929 cell growth rate and subside tumor tissue of transplanted tumors mice. Our team is the first research about DDW in China. The result shows that DDW inhibit the proliferation of lung cancer cell in vitro, while nothing to the natural cell It may caused by DDW which stops tumor cell in s period and promotes apoptosis of tumor cell, then DDW can play an important role on anti-tumor properties in vivo and vitro. Furthermore, DDW can also increase the level of some oxidation indicators in tissues like heart, liver, brain and blood, it means DDW improves the effect of antioxidation effectively. Base on these results, this research wants to explore the immunomodulating effect of DDW to further explain its anti-tumor properties and provide the laboratory data for secondary cancer treatment.2. MethodsAll of the mice(ICR,male,18-22g) are divided into 2 groups, one is the experimental group which accepted DDW as the water, the other is the control group. After 40 days, these mice will be cervical dislocate and measure the following immune parameters.(1) Non-specific immuneIt includes the measurement of Thymus index and Spleen index.(2) Humoral immuneIt includes the measurement of IgM, IgG in blood by using ELISA.(3) Cell-mediated immunityThe proliferation of Spleen lymphocytes, the activity of NK cell,CD4,CD4+/CD8+and dinitrofluorobenzeneinduced delayed type hypersensitivity test(DTH) are employed to evaluate the Cell-mediated immunity effect of DDW.(4) Pathological changes in lymphoid organ and evaluation of physiological characteristics On two groups of mice,spleen and thymus were stained and observed pathological changes by microscopic. Evaluate their physiological characteristics.3. Result(1) Non-specific immuneDDW can increase the level of Spleen index (3.93±0.35)vs(3.45±0.28)(P< 0.01),while Thymus index has nothing changed.(2) Humoral immuneDDW can increase the level of IgM in blood(1.10±0.10)vs(0.97±0.05)(P< 0.01),but never change the content of IgG.(3) Cell-mediated immunityTo explore the Cell-mediated immunity property, we make those following experiments like The proliferation of Spleen lymphocytes, the activity of NK cell, CD4, CD4+/CD8+and dinitrofluorobenzeneinduced delayed type hypersensitivity test (DTH).Those results show that DDW can significantly increase the number of CD4, CD4/CD8 and at the same time reduce the CD8's content, while other data has nothing changed.(4) Pathological changes in lymphoid organ and evaluation of physiological characteristicsBy HE staining,pathological reading shows that subjects with low deuterium water on the material the spleen and thymus in mice has no effect. Physiological characteristics of the control group has no difference.4. ConclusionDDW improves non-specific and specific immunologic function of normal mice.
