Higher fungi belongs to the very productive biologically source which produces a large and diverse varities of secondary metabolites. And these are very impotant for the searching work of some medicine and agriculture chemical leading compounds. In order to find some leading compounds and structures with bioactivities, the chemical constituents and bioactivities of the two higher fungi species of Lactarius vellereus (Fr.) Fr. and Suillus granulatus (L.:Fr) O. Kuntce have been investigated. And a review on both sructures and bioactivitise of the sesquiterpenes isolated from the higher fungus in the last decade is presented in the thesis.First, from the ethanol extract of the two higher fungi, thirteen compounds have been isolated by means of chromatographic methods. Twelve compounds are elucidated on basis of spectral(MS, ID & 2D-NMR, IR, UV), X-ray and chemical methods, including six sesquiterpenes, two steroids, one aromatic compounds, D-allitol,3-pyridinecarboxylic acid and Leptosphaepin that has not been isolated from higher fungus, only one novel compound, named Velleratretraol. Now, there is one cerebroside being elucidated and it may be a new compound by its MS.Second, anti-HIV, antitumor, antioxidant and antifungus of these compounds are tested. In anti-HIV, the CC50, EC50 and TI of LY-1 are above 200μg/mL,90.97μg/mL and above 2.2; the CC50, EC50 and TI of LY-2 are 82.53μg/mL,61.66μg/mL and 1.3; the CC50, EC50 and TI of LY-16 are 80.05μg/mL,32.27μg/mL and 2.5; the CC50, EC50 and TI of LY-18 are 136.47μg/mL,67.96μg/mL and 2.0; the CC50, EC50 and TI of LY-19 are above 200μg/mL, 51.71μg/mL and above 3.9; the CC50, EC50 and TI of LY-26 are 89.06μg/mL,58.05μg/mL and 1.5. In antitumor, the IC50 of LY-2 and LY-26 are 35.44μg/mL and 37.89μg/mL after 96 hours. The compounds tested have no bioactivities in antioxidant and antifunfus.
