Platinum-based antitumor drugs have been predominant place in cancer treatment. Mechanism of action and development of platinum antitumor complex were reviewed in this article. It is generally accepted that Pt(â…¡) complexes have enormous potential in anticancer, Pt(â…£) compounds must be reduced to Pt(â…¡) metabolites before binding to DNA. In the present investigation, the Pt(â…£) is substituted by glycine and N-methylglycine using the conventional spectrophotometry in the acid medium, just reflects the anticancer potential of these Pt(â…£) complexes. Based on theoretical calculation and experimental results, probable reaction mechanisms of substitution are proposed, and the rate equation derived from the mechanism can explain all the experimental phenomena.Gold compounds also have anti-cancer and anti-AIDS activity in recent clinical application. Mechanism of action and development of gold anti-cancer complex were reviewed in this article. Both Au(â…¢) and Au(â… ) have enormous potential as anticancer agents. The Au(â…¢) is substituted by L-Glutamine in the highly acid medium. And its reduction by Salicylic acid and Sulfosalicylic acid has been investigated in the lowly acid medium using the conventional spectrophotometry. Based on theoretical calculation and experimental results, probable reaction mechanisms are proposed, and the rate equation derived from the mechanism can explain all the experimental phenomena.
