Effect Of Calcitriol Combinating Ddp On The HEC-1-A Transplantation Tumor Of Athymic Mouse

Objective: To investigate the effect and mechanism of calcitriol combinating DDP on the HEC-1-A transplantation tumor of athymic mouse, and provide a new way for the treatment on endometrial cancer.Methods: To culture the human endometrial cancer(HEC-1-A) in vitro. The HEC-1-A were injected into the subcutaneouly of athymic mouse back. Divide the athymic mouse carring tumor into four groups,control group,calcitriol group,DDP group and combinating therapy group. Observe the growth condition,measure the volume and weight of tumor, detect the expression of Bax mRNA and Bcl-2 mRNA by RT-PCR,calculate the apoptosis rate of tumor cells by Annexin V-FITC/PI double staining with flow cytometry.Result: The change of body weight is same between calcitriol group and control group(P>0.05),but the difference between DDP/combinating therapy and control is significant(P<0.05).The difference of transplantation tumor volume between three therapy groups and control group is significant(P<0.05). The restraining tumor rate of three groups is 54.85%(DDP group),52.39%(calcitriol group),60.28% (combinating group).The apoptosis rate of the control group,DDP group,calcitriol group and combinating group are (1.45±0.23)%,(25.62±1.16)%,(22.52±1.25)% and(31.59±1.93)%, the difference are significant between two of them(P<0.05). The expression of Bcl-2 mRNA descent in order of control group,DDP group,calcitriol group and combinating group, but upgrade in order with Bax mRNA. The difference between three therapy groups and control group are significant(P<0.05).Conclusion: The calcitriol can induce the HEC-1-A transplantation tumor cells apoptosis by the way of Bax and Bcl-2. Calcitriol combinating DDP can enhance the curative effect and relieve the side effect of DDP.
