PartⅠVariation of the expression of phosphorylated Akt and its significance in the endometrium of controlled ovarian hyper-stimulation mouse during the period of "implantation window"Objective To investigate the expression of pAKt and LIF in the endometrium of the mice in natural cycle group and that of controlled ovarian hyper-stimulation group during the period of implantation,explore the influence of Controlled Ovarian Hyper-stimulation(COH) on uterine endometrium receptivity and the possible relation between pAKt and uterine endometrium receptivity.Methods 48 female mice were randomly divided into 2 groups,the natural cycle as control group,COH group as test goup.Mice in the COH group were intraperitoneally injected with GnRHa each day at 9Am during luteal phase for 9 days,on the 9th day PMSG was injected additionally,48 hours later,hCG was injected..Nothing was given to mice in control group and the estrus cycle was observed only.Mice in the COH group were mated with male mice at ratio of 2:1 on the night of the day injecting hCG,mice in natural cycle were mated with male mice at same ratio on the night of the day of their ovulation.The next morning,vaginal plugs were observed,pregnance day 1 was determincd on the vaginal plug appeared,three pregnant mice in each group were labelled on each day as planned,and lasting for 6 days,the pregnant and non-pregnant female mice were all labeled.The mice of the 4th pregnant day were killed by cervical dislocation at the time of 23:00-24:00,then uteruses were collected,other mice were killed at 9Am on the day needed in the same way,the blastocyst in the site of imbed needed to be removed from the mice's uteruses of the fifth and sixth pregnant day.Immunohistochemistry assay was used in all samples to determine the tissue location of pAKt and LIF and their semi-quantitative expression.Results(1) LIF located mainly at endometrial glandular epithelial and luminal epithelial cells' cytoplasm of the mice during the implantation windows of the two group.(2) The levels of pAKt increased markedly in stromal cells and cytoplasm showed strong expression in all mice' endometrium.(3) Endometrial LIF protein semi-quantitative expression of the mice in the two groups revealed as followed,compared with control group,LIF protein were significantly decreased in the COH group(P<0.01).The expression of LIF protein in the pregnant group were significantly higher than that of non-pregnant group(P<0.001). (4) Endometrial pAKt protein semi-quantitative expression of the mice in the two groups revealed as followed,pAKt protein expression were significantly decreased in the COH group compared with control group(P<0.001),the expression of pAKt protein in the pregnant group were significantly lower than that of non-pregnant group(P<0.001).Conclusions(1) COH could decrease the expression of pAKt and LIF during the implantation windows of the mice.(2)The variations of the expression of pAKt and LIF were relatively synchronous,which suggest that pAKt may be a candidate marker of uterine receptivity. Objective To study on variation of the expression of pAKt and LIF in the endometrium of women with unexplained infertility and patients with obstruction of two fallopian tube, and on the possible relation between the variation of pAKt expression in the endometrium and endometrium receptivity of unexplained infertile women.Methods 18 samples of the endometrium in midterm of the luteum from the women with primary and unexplained infertility were collected as test group,and 30 samples of the endometrium in midterm of the luteum from the patients with secondary infertility and two fallopian tube obstruction were also collected as control group.SP immunohistochemistry assay was used to determine the tissue location of pAKt and LIF and their semi-quantitative expression of the endometrium in the two groups.Results(1) LIF expression were located in endometrial glandular epithelial cells and luminal epithelial cells of the two group.(2) The expression of pAKt markedly located in stromal cells and cytoplasm showed strong expression in the patients' endometrium of the two groups.(3) Endometrial LIF and pAKt protein semi-quantitative expression in the two groups(SP immunohistochemistry assay)revealed as followed,compared with the patients with two fallopian tube obstruction,LIF and pAKt protein expression were both significantly decreased in the unexplained infertile women(P<0.05).(4) The expressions of LIF and pAKt protein in the pregnant patients after IVF/ICSI in the two groups were both significantly higher than those of the non-pregnant patients(P<0.01).Conclusions(1) Decreasing of endometrium receptivity is one of the infertile causes of women with unexplained infertility.(2) The variations of the expression of pAKt and LIF were relatively synchronous in the endometrium of the infertile patients in two groups, which indicate that decreased expression of pAKt in the endometrium during "implantation window" may be one of the causes of infertile women's decreased endometrium receptivity.
