High performance liquid chromatography (HPLC) plays an important role in pharmaceutical analysis at present. Column is the heart of the HPLC, so the bulking agent of the column has an influence on the efficiency of the separation. Because of the advantages of good permeability and simple preparation, the monolithic column has been applied widely in HPLC for analysis.In this paper, a review concerned with the development and application of monolithic column was introduced firstly.A monolithic column was prepared using vinyl ester resin both as the monomer and the cross-linking agent. Using this column, nisoldipine in human plasma samples could be on-line enriched and meanwhile the protein could be eluted. Moreover, twelve-hour pharmacokinetic profile of nisoldipine in mouse after oral administration was investigated. Results indicated that this method could be applied for therapeutic drug monitoring of nisoldipine and it provided a simple and fast method for assaying this drug in plasma sample.Molecular imprinted polymer of mycophenolate mofetil (MMF) was synthesized using vinyl ester resin as the cross-linking agent and methcrylic acid as the monomer. HPLC was used to study the molecular recognition performance, to evaluate the binding performance of MIPs for template and its cleavage product Mycophenolic acid (MPA). For chromatographic analysis, the effects of mobile phase composition, flow rate, and mobile phase PH were investigated on the separation. The adsorption isotherms of MMF on the imprinting stationary phase were determined using rectangular pulse frontal analysis. In addition, a solid-phase extraction column packed with MMF-imprinted polymers was used to enrich the MMF.
