Objective:To clone the homologous sequences of Human Testis Development Related Gene 1 (TDRG1) from Macaca mulatta and to explore the potential animal models for further researching the function of TDRG1Methods:The nucleotide sequence of TDRG1 was used as a query sequence to search for the rhesus macaque genome databases by BLASTN program to obtain the homologous sequences. The homologous sequences of TDRG1 were then confirmed by RT-PCR. Northern blot,Real-time PCR and Immunohistochemistry were used to detect the expression level of TDRG1 in rhesus macaque testicle tissues and different tissues of the rhesus macaque.Results:Bioinformatics analysis indicated that homologous sequences of TDRG1 were found in the rhesus macaque genome databases, and the percent similarity was 94%. The homologous sequence in rhesus macaque testicle was confirmed by RT-PCR. Northern Blot and Real-time PCR detected that the homologous gene of TDRG1 exclusively expressed in rhesus macaque testicle, not in kidney, bladder, liver, and so on. Immunohistochemistry showed that anti-TDRG1 Monoclonal Antibody can specifically combine the homologous protein in rhesus macaque testicle, but no homologous protein expressed in kidney,bladder,liver,and so on.Conclusion:There are homologous genes of TDRG1 in rhesus macaque testes. This study provide theoretical and experimental basis for developing TDRG1 transgenic or knockout animal models.
