The Extract Of Shiraia Bambusicola And Its Effects On The TNF-α Pathway In Rheumatoid Arthritis

Objective:TNF-αinduced L929 cell line was used as a model to explore the mechanism in the treatment of rheumatoid arthritistr with tranditional Chinese medicine Shiraia bambusicola. The effect of its extract on L929 cells growth and apoptosis, and the NF-κB protein expression changes in L929 cells line were especially discussed. The mechanism of Shiraia bambusicola's extract interfering with TNF-αsignal transduction is analysised and the results obtained from this research provide a theoretical basis in clinical treatment of rheumatoid arthritis.Methods:(1) Soxhlet extraction was applied to extract the effective components of Shiraia bambusicola (2) Morphological changes of the cells were observed by microscope. The cell growth inhibition was measured by SRB assay. The rate of apoptosis was analysed by FCM. Hoechst staining analysis was applied to observe the possible apoptosis. Western blotting was applied to detect the expression of NF-κB P65 protein in L929 cells.Results:(1) Crimson unguent is obtained in the research, the component analysis supplies that the unguent may contain Perylenequi-nonoid compounds; (2) Shiraia bambusicola extract are capable of abating the morphological changes of L929 cell line which are caused by TNF-α. The SRB assay suggests Shiraia bambusicola extract can decrease L929 cell growth inhibition caused by TNF-α, and that is a concentration-dependent effect. Hocehst staining and Flow cytometry suggests Shiraia bambusicola extract can reduce the rate of L929 cell apoptosis which is affected TNF-α, and that is also a concentration-dependent effect.Conclusion:Shiraia bambusicola extract is capable of down regulating the expression of NF-KB p65 protein in L 929 cell line which is induced by TNF-α. Shiraia bambusicola affect TNF-αpathway and inhibit of NF-κB activity may be play a key role in treatment of rheumatoid arthritis.
