| Objective To study the impacts of Intense Pulsed Light(intense pulsed light, IPL)on transforming growth factor beta-1 secretion in cultured human fibroblasts and the signaling mechanism in the process.Methods The callan foreskin fibroblasts were cultured and then divided into two groups. The first group,the IPL treatment group, was irradiated with IPL with fluences of 0 J/cm2 (negative control),10 J/cm2, 18 J/cm2,27 J/cm2,36 J/cm2,36 J/cm2×2(irradiated twice with IPL with fluences of 36 J/cm2).The second group, IPL+inhibitor group, including the IPL (control), IPL+SP600125(JNK inhibitor), IPL+SB203580(P38 inhibitor),was irradiated with IPL with fluences of 36 J/cm2 two hours after adding the inhibitor SP600125 and SB203580. TGF-β1 in cultural supernatants of the two groups was evaluated using enzyme-linked immunosorbent assay and p-JNK (phosphorylated JNK) and p-P38 (phosphorylation P38) in the IPL treatment group were evaluated using western blot.ResultsThe concentrations of TGF-β1 in cultural supernatants of two groups①The concentrations of TGF-β1 in cultural supernatants of the IPL treatment group:the concentrations of the TGF-β1 in cultural supernatants of 10 J/cm2,18 J/cm2 and 27 J/cm2, reduced significantly compared with the negative control (P<0.05), in 36 J/cm2,also reduced(P>0.05),respectively, while in 36 J/cm2×2, increased (P< 0.05).②The concentrations of TGF-β1 in cultural supernatants of the IPL pluses inhibitor group:the concentration of the TGF-β1 in cultural supernatants of IPL+SP600125 reduced compared with the control,while in IPL+SB203580, increased(P<0.05).2. The expression of JNK and P38 in IPL treatment group:the erpression of JNK and P38 first increased and then decreased with the fluences increasing. The strongest expression in 10 J/cm2 Both of them began to weaken in 18 J/cm2, then decreased significantly.Conclusion 1.Intense pulsed light could suppress the secretion of TGF-β1 at the lower fluences,while could promote it at higher fluence.2.Protein kinases JNK,p38 may played opposite roles in the process of IPL affecting the secretion of TGF-β1, in which JNK promoted its secretion and P38 inhibited its secretion. |
