Object:Chinese herbal medicine theory is the main content of traditional Chinese medicine, in which cold and heat effects of the medicine is the core theory. The traditional Chinese medicine herbs are almost from the experience more than theory. The current study of the modernization of traditional Chinese medicine prefers to studying the monomer constituents under the guidance of natural substances of plant chemistry and chemistry theory based on Western medicine, and less involves with the characteristics of traditional Chinese medicine herbs. As an emerging discipline of systems biology, the overall system of the use of metabonomics technology consisted with the traditional theory of TCM. The project intends to choose cold and hot different natures based on traditional Chinese medicine classification, and study the biological changes in metabolites caused by different Chinese herbal medicine. According to these changes, the study intends to look for the relationship between drug potency and the metabolic products, select the characteristic metabolites of different potency,and establish the doscriminant model which can be used to identify the characteristics of traditional Chinese medicine herbs.Method:1. Serum collection of Chinese medicine:select 19 single drug particles of Chinese herbal medicines, depending on the medication is divided into four groups, heat, cold,, each containing 10 different herbs (including 9 herbs in hot group), while the establishment of solvent control group and blank control group. SD rats will be kept under the same conditions for six days and then collect the serum form the orbital venous plexus blood as a control sample of the former administration. After taking blood, f or a single drug gavage 7 days, daily one time, the dose= commonly used for the human dose/person standard body weight * 20/g. In the seventh day, two hours after administration, collect the serum form the orbital venous plexus blood again.2. Metabolomics analysis:After the pretreatment of the serum, the sample will be tested based on HPLC-Q/TOF MS (high performance liquid chromatography/time of flight mass spectrometry) and be analyzed for all components of metabolomics.3. The raw data through the R software, t-test and PLS-DA analysis, get relevant information of various metabolites, the establishment of Chinese herbal medicine-based metabonomics analysis model.4. P value less than 0.05 as a standard to determine the comparison between different groups of metabolites derived from differences.Results:1. By HPLC-TOF/MS, it can detect the different potency drug-induced changes in body's metabolism. Of which are drug-specific changes in hot group plasma metabolites 41 species, and cold drug-specific changes in plasma metabolites are 30 species.2. By PLS-DA method can distinguish between hot and cold potency differences in the initial establishment of the model, explained 85.3% of classified information, the model predicted rate of 61.8%.Conclusion:1. The different potency of drug metabolites can be analysised in the different drugs, and can find the specific part of the potential biomarkers.2. In this study, differences in metabolites found need to be further confirmation by MS/MS.3. Metabolites are obviously different between cold and hot effects of the medicine, and the difference models between the two herbs can be initially established.
