Objective:Observe the influence of cold and heat TCM on normal rats to explore the relationship between the cold and heat property of TCM and its efficacy; collect HPLC spectrogram of cold and heat TCM to explore the relationship between the cold and heat property of TCM and its material composition from the perspective of integrated chemical information of TCM; observe the effect differences of treating rats with deficiency-heat syndrome or deficiency-cold syndrome by using different extract from the same cold or heat medicine to explore the relationship among the cold and heat property of TCM, its efficacy and its material composition.Methods:Select the typical sample of cold and heat TCM. The sample is recognized by the authority and people raise no objection to its cold and heat property both in the ancient and modern times. Collect drug effects indicator data of normal rats after administration, determine the correlation of the individual indexes and cold and heat property of medicine by single factor analysis, get the full gene expression profiles data of the rat liver by using gene chips to screen for genes with significantly changed expression, check its function and expression pathways, and use Bayesian belief network to conduct the data mining on the experimental data; from the HPLC spectrogram database of typical cold and heat TCM, after treatment of compression, dimensionality reduction and squelch, extract96marks of medicine nature to represent material composition of cold and heat medicine for treating cold and heat syndrome, and select the PLS-DA model which has the highest whole recognition rate for the marks of medicine nature to test its accuracy rate of back substitution and correct discrimination rate of cold and heat property of TCM. Base on what has been talked above use Bayesian belief network to conduct data mining on the correlation between the cold-and-heat property and the marks of TCM property; extract the main cold and heat material composition from the accumulation parts of cold and heat marks of cold medicine anemarrhena and heat medicine zingiberis. Use anemarrhena extract and decoction to treat rats with deficiency-heat syndrome and use zingiberis extract and decoction to treat rats with deficiency-cold syndrome. Collect the rats drug effects indicator data to conduct single factor analysis.Results:Establish respectively the "Bayesian belief network of the correlation between normal rats drug effects indicator and cold and heat property of medicine","Bayesian belief network of the association and relationship between full gene expression change of normal rats liver and cold and heat property of medicine" and "Bayesian belief network of the association and relationship between cold and heat property of TCM and marks of medicine nature". Graphically display the causal relationship between cold and heat property of TCM and drug effects indicator, the causal relationship between cold and heat property of TCM and the obvious change of gene expression and its action way, as well as the compatibility pattern and combinations path of cold and heat property of TCM and its medicine nature mark. The three Bayesian belief networks can forecast the property of TCM through different index variables; Meanwhile, the result of the network topology analysis indicates that there is a clear marker structure of cold and heat property and a clear modular structure can be seen. This means that the heat substances gather together to form a special composition to match the pattern and pathway; some heat markers are mixed in the module of cold substances, but these markers are non-critical network nodes and they are in the periphery of the network. Thus, the modularity and pathways of its pattern matched are comparatively obvious.Conclusion:The TCM with cold and heat property not only can correct the excess cold or excess heat of the organism in a pathological state but also can cause the cold-and-heat effect in a normal organism. The material composition with cold or heat property exists in any TCM. There is something in common in the material compositions of TCM with the same property on terms of the compatibility pattern and combinations path. The overall property of the TCM can be displayed in the combination of all its material compositions. Some material composition or some single ingredient can not characterize the overall pharmacodynamic effect of certain TCM. A single TCM is also a "compound medicine" and only the combinations of quality and quantity of the different ingredients in it may characterize the "cold" or "heat" property of the overall pharmacodynamic effect. There is a big difference in treatment of the same disease for the main cold material composition and the main heat material composition in the same medicine. The main heat material composition of the heat medicine and the main cold material composition of the cold medicine have the equal efficacy of decoction of TCM. The main cold ingredients of heat medicine as well as the main heat ingredients of cold medicine have little medicine efficacy. Meanwhile, the Bayesian belief network is an effective data mining method for the correlation research of the property, efficacy and material composition of TCM, which can combine the prior knowledge and observation data to deduce the causal relationship and the condition correlation among the variables of network nodes through probability computation, making the graphical visualization of the results.
