In recent decades, with the rapid socio-economic development, the people's living standards and the quality of life have improved continuously, and the life style has changed greatly. At the same time, manual labor and physical activity also decreased, which resulting in a negative balance of energy metabolism on the health of people and bringing great risks to people's health including obesity, hyperlipidemia, diabetes mellitus, coronary heart disease, metabolic syndrome and other metabolic diseases. Because these metabolic diseases have a serious threat to human health and the society, so to investigate their molecular biology mechanisms and the means of prevention have turned to be very important and popular. Sports as a part of healthy lifestyles of human being has played an important role in improving the body's metabolisms, preventing and mitigating metabolic diseases. Traditional views considered that aerobic exercise is one of the effective means of preventing obesity and weight-loss. Research shows that regular resistance exercise can increase muscle strength and size, improve the lean body mass and basal metabolic rate, promote muscle fat metabolism, and increase insulin activity in skeletal muscle. So we think that resistant exercise combined with aerobic training might be able to better promote the improvement of muscle metabolism, improve muscle function, regulate lipid metabolism.In this paper, we will discuss the 3 different ways of exercises acting on 2-month-old C57BL/J6 mice skeletal muscle, to observe the changes of mRNA expression level of ATGL, HSL and ADRP, so as to explore the levels of adaptive changes in IMTG affacted by different ways of exercises, while put forward the optimal mode of exercise in promoting the metabolism of the fat and the skeletal muscle. And to establish a feasible method of preventing fat metabolic disorders for human being.Experimental method:This study selected 32 2-month-old C57BL/J6 male mice as the objects of study, according to the body weight, we randomly divided them into sedentary control group (C), endurance exercise group (E), resistance training group (R), endurance exercise and resistance training group (E+R),8 mices in each group. Running on the treadmill as the mode exercise of endurance exercise, while climbing the ladder as the mode exercise of resistance training. After 29w exercise, the mices were killed within 48h after the last exercise, then quickly take the posterior right quadriceps of mice, weight and number the samples, then put into Eppendorf tubes. Following the samples were frozen in liquid nitrogen, and transferred to-80 ℃refrigerator.To determine the mRNA expression changes of ATGL, HSL, ADRP by the means of real-time PCR, presenting by mean±standard deviation (Mean±SD). Data were processed by the statistical analysis software SPSS 18.0. the differences between the groups using independent sample T test analysis, P<0.05 as significant difference, P<0.01 as highly significant difference.The results:1, After 28 weeks regular exercise, the body weight of mice have increased, but the rate of increasing is reduced by the time. The sedentary control group has the largest change range, and endurance exercise group has the smallest.2, Compared with the control group, after 28 weeks of regular exercise, the weight of quadricep divide body weight ratio (V/W) had significantly increased (P<0.01) of endurance exercise group and resistance exercise group and mixed group.3, Compared with the control group, three kinds of exercise intervention, aerobic exercise can increase the HSL activity significantly; While resistance exercise made the HSL activity reduced in a certain extent, and the effect of mixed exercise on HSL activity has not be detected.4, Compared with the control group, three kinds of exercise intervention, in aerobic exercise group and mixed group, FFA were significantly increased with significant difference (P<0.01); Resistance exercise group has not different (P>0.05).But there is significant difference between aerobic exercise group and resistance exercise group (P<0.01);There is aslo significant difference between mixed group and resistance exercise group (P <0.05);While there is no difference between aerobic exercise group and mixed group(P> 0.05);5, Compared with the control group, three kinds of exercise intervention, the ATGL mRNA expression of aerobic exercise group and resistance exercise group have significantly increased (P<0.01); mixed group has no difference (P>0.05); Compared with mixed group, the ATGL mRNA expression of aerobic exercise group and resistance exercise group have significantly increased (P<0.01); Compared with resistance exercise group, the ATGL mRNA expression of aerobic exercise group has significant difference (P<0.01).6, Compared with the control group, three kinds of exercise intervention, the HSL mRNA expression of aerobic exercise group and resistance exercise group have significantly increased (P<0.01); mixed group has no difference (P>0.05); Compared with mixed group, the HSL mRNA expression of aerobic exercise group and resistance exercise group have significantly increased (P<0.01); Compared with resistance exercise group, the HSL mRNA expression of aerobic exercise group has significant difference (P<0.01). 7, Compared with the control group, three kinds of exercise intervention, the ADRP mRNA expression of aerobic exercise group and resistance exercise group have significantly increased (P<0.01);But mixed group has significantly decreasing (P<0.01); Compared with mixed group, the the ADRP mRNA expression of aerobic exercise group and resistance exercise group have significantly increased (P<0.01); Compared with resistance exercise group, the ADRP mRNA expression of aerobic exercise group has no significant difference (P >0.05).Conclusion:4.1 Aerobic exercise can increase ATGL, HSL, ADRP mRNA expression significantly,and can increase the activity of HSL significantly, muscle mass against body weight was significantly increased, simultaneously the body weihgt was increased significantly,all of above suggested that lipolysis was enhanced,which may promote IMTG metabolism.The effect of aerobic exercise was stonger than resistance exercise.4.2 Resistance exercise can increase ATGL, HSL, ADRP mRNA expression significantly, and muscle mass against body weight was significantly increased,but the body weight was decreased significantly,with the activity of HSL decreased significantly,suggested that resistance exercise aslo has certain stimulative effect to ATGL, HSL, ADRP mRNA expression,but the effect was weaker than aerobic exercise.4.3 Mixed exercise had no significant effects on ATGL, HSL mRNA expression,but it can make ADRP mRNA expression decrease significantly, the activity of HSL had no significant changes, Maybe because endurance exercise in mixed sports and lack of resistance exercise in physical activity that not have sufficient stimulus on the fat metabolism genes or enzymes to generate.
