With the advent of aging society in China, the structure and function of many tissues and organs of the elderly population will decline, one of the obvious symbols is skeletal muscle ageing. Skeletal muscle is the most important movement organ in human body, in the process of ageing, decline of the skeletal muscle function is the important symbol of aging. There is a relationship between aging and further decrease of strength, quality of muscle, usually due to this change in the muscle is called Sarcopenia. Due to loss of muscle activity, which indicates that there will be more body deformity and mortality. And these are in connection with poor quality of life, social needs and health care needs. Based on the serious threat of Sarcopenia, it is important to in-depth analysis the molecular mechanisms and preventive means of Sarcopenia.Sports is an integral part of human healthy lifestyle, it plays an important role in the process of prevention and mitigation of the muscle aging. Exercise, In particular, regular resistance exercise,some extent,to reverse the aging of muscles, delay the further development of Sarcopenia, and and increase muscle strength, size, and non-adipose tissue weight, and promote the muscle health of the elderly. A variety of exercise has a different effect on the occurrence of skeletal muscle, however, there is few research about different forms of movement of the animal or human muscle-derived muscle hypertrophy ability response and molecular mechanisms, and cell signaling pathway changes and the degree of influence by the changes in training patterns associated with muscle hypertrophy are poorly understood.In this paper, we observed ERK1/2, mTOR, p70S6K mRNA expression changes in the skeletal muscle of 2-month-old C57 male mice by three different kinds of movements after 28 weeks of intervention, in order to examine the effects of exercise on skeletal muscle protein synthesis and the the effects on different muscle groups that degree of change occurs. At the same time for the selection of effective mode to promote skeletal muscle hypertrophy and delay aging, and provide feasible methods and theoretical basis to develop exercise prescription that could significantly enhance muscle strength..Experimental Methods: This study selected 32 2-month-old male C57 mice,and randomly divided into sedentary control group (C), endurance exercise group (E), weight-bearing ladder group (R), mixed sports group (E+R), n=8. Running on the selected object and the weight-bearing ladder used respectively as the endurance and resistance exercise mode, a period of 28 weeks. Application of RT-PCR detection method to determine ERK1/2, mTOR, p70S6K mRNA expression.Experimental results:1, Exercise significantly reduces the weight of mice.2, Compared with a control group, endurance, resistance groups and mixed group of mice the gastrocnemius wet weight to body weight ratio (G/W) has a very significant rise; quadriceps wet weight to body weight ratio (V/W) and similar.3, Resistance group mice gastrocnemius ERK1/2 mRNA than other three groups are very significant increase; quadriceps in resistance group, but a mixed set of expression in the control group and endurance group appears very significant decline.4, Resistance within the gastrocnemius muscle of mice the expression of mTOR mRNA than the other three groups, the increases were highly significant; quadriceps resistance group within the expression of mTOR mRNA than the control group and mixed group is concerned, was very increased significantly. Endurance group was relatively mixed group increased significantly.5, Resistance group mice gastrocnemius p70S6K mRNA than other three groups are very significant increase. Endurance group relative increase in mixed groups are significant. Mixed groups expressed a control group is extremely significant decline; quadriceps in resistance groups p70S6K mRNA a control group is a significant increase in its more endurance group and mixed groups increased very significantly.6, The group of mice gastrocnemius ERK1/2 mRNA its corresponding group quadriceps ERK1/2 within the mRNA, resistance group gastrocnemius expression is significantly higher than quadriceps in expression; mTOR mRNA, the resistance group gastrocnemius expressed very significantly higher than the quadriceps in expression; resistance group p70S6K mRNA expression and mTOR mRNA similar, but other groups within the gastrocnemius expression are significantly higher than the quadriceps in expression. Conclusion:1, Exercise affected the body weight of mice in the control group gained weight at most, generally due to increased fat; while the smallest changes in endurance exercise on body weight, usually caused due to fat consumption more.2, Three different forms of movements so that the proportion of mice gastrocnemius and quadriceps to increase, the increase in endurance exercise group may be due to a decrease in the relative fat; resistance exercise group may be due to the increase in muscle mass itself the increase of; mixed exercise group may be due to the increase in the relative reduction of fat or muscle due to the increase in their quality.3, Three kinds of exercise mode, resistance exercise to gastrocnemius and quadriceps in ERK1/2, mTOR, p70S6K mRNA expression were significantly increased, it suggested resistance movement pattern so far is effective to activate muscle hypertrophy, muscle mass increase in signal transduction pathway as well as the delay Sarcopenia movement occurred in the preferred or best way.4, Endurance sports model within the gastrocnemius or quadriceps ERK1/2, mTOR, p70S6K mRNA expression was not significantly impact statement resistance of skeletal muscle aging, increase muscle volume, endurance exercise on the form of little importance, there is no real desirability.5, Mixed movement pattern to gastrocnemius and quadriceps in ERK1/2, mTOR, p70S6K mRNA expression occurred decrease, indicating the mode of endurance exercise inhibits the effect of resistance exercise, suggesting that increase in muscle mass and delaying muscle aging are trained to properly handle the relationship between these two kinds of movement pattern, pay attention to their time intervals and the respective exercise intensity.6, Gastrocnemius muscle with ERK1/2, mTOR, p70S6K mRNA expression is always greater than the expression of the quadriceps, the expression that within to typeâ…¡of muscle fibers faster in the protein synthesis of gene expression is always above typeâ… andâ…¡muscle fibers, the proportion is mixed increased the quality of skeletal muscle movement clew occurs mainly in the biological effects of typeâ…¡muscle.
