Objective:To evaluate the effects of COX-2 inhibitor Celecoxib on the proliferation,migration,adhesion and invas-ion of MG-63 and U2-OS Osteosarcoma cell lines,explore the molecule mechanisms of these effects.Method:MTT assay, scratch wound assay, adhesive assay and Transwell chamber in vitro invasion assay were employed to detect the change of the proliferation, migration, adhesion and invasion ability of the cell lines which treated by different concentration(25,50,100μmol/L)of celecoxib for 24 hours,flow cytometry and Western blot were used to detect the change of CD44 and MMP-9 expression after the treatment.Result:The proliferation,migration,adhesion and invasion ability of the two cell lines were significantly inhibited after the treatment of different concentration's celecoxib,the expression of CD44 and MMP-9 were downregulat-ed,all these effects presented a dose-dependent manner, these inhibition effects were more obvious in MG-63 cell line which expressed a higher level of COX-2.Conclusion:The selective COX-2 inhibitor celecoxib can inhibit the proliferation,migration,adhesion and invasion of human Osteosarcoma cell lines through downregulating the expression of CD44 and MMP-9,which was COX-2-dependent.
