Purpose:osteosarcoma MG63 cells cultured in vitro were our object of study,the Pyropheophorbide-? methyl ester mediated photodynamic therapy was used to treat osteosarcoma MG63 cells,to investigate the effect and its possible mechanism of pyropheophorbide-a methyl ester-mediated photodynamic therapy(MPPa-PDT)on the invasion and migration abilities of osteo sarcoma MG63 cells.To provide experimental theoretical basis for MPPa-PDT in distant metastasis inhibition and clinical treatment of osteosarcoma.Methods:Human osteosarcoma MG63 cells in logarithmic growth phase were divided into 4 groups: blank control group(control group),the MPPa treatment group(MPPa group),the light irradiation group(LED group),and MPPa-PDT treatment group(MPPa-PDT group).Osteosarcoma MG63 cells was treated by MPPa-PDT,and then cell proliferation were subjected to CCK-8 assay at different point,and cell migration and invasion were detected by Wound healing assay and Transwell chamber model experiments,the expression levels of E-cadherin(E-cad)and matrix metalloproteinase(MMP)-2,-9 protein were detected by western blot.Results:After osteosarcoma MG63 cells were treated by MPPa-PDT,cell proliferation of MPPa-PDT group had no significant change than the control group,MPPa group,LED group;Compared with the control group,MPPa group,LED group,osteosarcoma MG 63 cells scratches healing,vitality migration and invasion abilities of the MPPa-PDT group were significantly decreased;Western blot test results were displayed that E-cadherin of the MPPa-PDT group E-cadherin was significantly increased than the other three groups,and matrix metalloproteinase MMP-2,MMP-9 expression was significantly lower than other groups.Conclusion:MPPa-PDT inhibited invasion and migration of human osteosarcoma MG63 cells;Upregulation of E-cad,down MMP-2,-9 expression may be one of the mechanisms.
