OBJECTIVE:To investigate the changes and mechanism of heart function and mocardial cells apoptosis after the transplantation of adipose tyissue-derived mesenchymal stem cells(ADMSCs) into rabbit models with myocardial infarction.METHODS:1.The adipose tissue-derived mesenchymal stem cells(ADMSCs) were isolated, cultured in vitro from rabbits and to marked by DAPI. Totally 30 Japanese big ears albino rabbits 5 months aged older. were selected selected and randomly divided into 3 groups:(sham-operation group,A;control group,B;ADMSCs transplantation group,C,(n=10);The rabbits in the two groups (B and C) were established into myocardial infarction models.2.The left antero decendant arteries (LAD) of rabbits were ligated so as to establish animal models of acute myocardial infarction (AMI). Then the marked ADMSCs were transplanted into rabbits in the ADMSCs transplantation group through epidium. Those in the control group were treated with Phosphate Buffered Saline (PBS) medium of the same volume. Thoracic cavity was closed routinely, and then cultured for 4 week successively.3.Before transplantation and 4 weeks after transplantation, all canines received ultrasonic cardiogram (UCG) test to determine left ventricular ejection fraction (LVEF),left ventricular end-diastolic diameter (LVEDD), left ventricular end-systolic diameter (LVESD). Four weeks later,rabbits were killed to obtain heart sample,and then the transplanted cells survival was observed. The apoptotic cardiomyocytes were measured with transmission electron microscopy and counted with in situ nick end labeling (TUNEL) method and light microscopy and left ventricular remodeling was measured.CONCLUSION:1.The left anterodecendant arteries (LAD) of rabbits were ligated so as to establish animal models of acute myocardial infarction (AMI).The model of myocardial infarction is reliable, reproducible, and Similar to the human condition,amenable to investigate other investigation.The therapy of implantation of autologous mesenehymal stem cells by Intramyocardial injection was effective, feasible and safe in ischemic heart disease.2.Transplanted ADMSCs can exist in host of rabbits.3.The transplantation of adipose tissue-derived mesenchymal stem cells(ADMSCs) can reduce apoptosis, reduce left ventricular remodeling and improve the heart function in rabbits with heart failure after acute myocardial infarction.
