ObjectiveTo investigate cytotrophoblast invasive ability and human umbilical artery smooth muscle cell apoptosis in the model of cytotrophoblast coculture with human umbilical artery smooth muscle cell induced by preeclampsia serum.Methodschorionic villous tissue of normal 6-8 week pregnancy were obtained. cytotrophoblast(CTB) and human umbilical artery smooth muscle cell (HUASMC) were collected by explant culture.construct the model of CTB coculture with HUASMC and incubated with normal or preeclampsia serum for 24 hours, as controls, CTB or HUASMC were monocultured and treated identically to the coculture situation. Analysis of the cytotrophoblast invasive ability,HUASMC activity and apoptosis,expressions of MMP-2,MMP-9,Fas,FaslmRNA and Fas protein were detected with Transwell invasion assay,CCK-8,flow cytometry,reverse transcription-polymerase chain reaction(RT-PCR) and Western blot.ResultsIn the model of CTB coculture with HUASMC,the preeclampsia serum can effectively decrease the cytotrophoblast invasive ability,increase HUASMC activity and decrease its apoptotic rate and decrease the expressions of MMP-2,MMP-9,Fas,FaslmRNA and Fas protein.Conclusionthe preeclampsia serum affect cytotrophoblast invasive ability in a model of CTB coculture with HUASMC maybe via regulae the expression of MMP-2 ,MMP-9, HUASMC apoptosis decrease may relate with the abnormal expression of Fas/Fasl.This data provide a rationale that the abnormal expression of MMP-2, MMP-9 and Fas/Fasl system may participate in the defective remodeling of the uterine spiral arteries is associated with pregnancies complicated by preeclampsia.
