Objective:This study aimed to investigate the presence of TIPE2(Tumor necrosis factor-a-induced protein.8like.2, TIPE2) and FasL in placenta among women with moderate preeclampsia, severe preeclampsia and normal pregnancies. We tested the hypothesis that the onset and development of preeclampsia are associated with TIPE2through Fas-induced apoptosis.Materials and Methods:We recruited25pregnant women diagnosed with preeclampsia (moderate preeclampsia, n=15and severe preeclampsia, n=10) and15women with normal pregnancies during July.2013and Mar.2014in Central south university xiangya second hospital. ALL patients were singleton pregnancy.TIPE2and FasL protein, TIPE2and FasL mRNA of maternal placenta were measured by Western blot and RT-PCR respectively. Statistic analyses were performed as well.Results:1. Expressions of TIPE2mRNA were decreased in preeclampsia patients as compared to controls (0.925±0.117), furthermore, TIPE2mRNA in severe preeclampsia was significantly lower than that in moderate preeclampsia (0.218±0.061vs.0.467±0.07) these three groups are statistically significant difference (P<0.001).2. Expressions of TIPE2protein were decreased in preeclampsia patients as compared to controls (0.286±0.056), furthermore, TIPE2protein in severe preeclampsia was significantly lower than that in moderate preeclampsia (0.068±0.023vs.0.146±0.030) these three groups are statistically significant difference (P<0.001).3. Expressions of FasL mRNA were decreased in preeclampsia patients as compared to controls (0.943±0.157), furthermore, FasL mRNA in severe preeclampsia was significantly lower than that in moderate preeclampsia (0.48±0.073vs.0.204±0.052) these three groups are statistically significant difference (P<0.001).4. Expressions of FasL protein were decreased in preeclampsia patients as compared to controls (0.262±0.075), furthermore, TIPE2protein in severe preeclampsia was significantly higher than that in moderate preeclampsia (0.065±0.022vs.0.163±0.029) these three groups are statistically significant difference (P<0.001).5. Expressions of TIPE2among three groups of cases were positively correlated with presences of FasL.Conclusions:1. Both TIPE2and FasL were expressed in human placental tissues, TIPE2levels in maternal placenta of preeclampsia were positively correlated with presences of FasL;2. Presences of TIPE2and FasL were reduced in preeclampsia patients compared to controls;3. Presences of TIPE2and FasL were negatively correlated with severity of diseases.
