| Objective To study von Willebrand factor's(vWF) effect on the proliferation,adhesion and migration in human colorectal cancer cell.Methods Cultured human colorectal cancer cell line SW480, detected by immunocytochemistry in SW480 cells the expression of vWF in vitro. With vWF antibody (vWFAb) treatment on SW480 cells, using an inverted microscope to examine cell morphology changes; Detected with MTT cell proliferation changes and extracellular matrix-Ⅳcollagen changes in adhesion capacity; the migration abilities of SW480 cells were assayed in Transwell.Results The human colorectal cancer cell line SW480 expressed vWF, staining occurred mainly in the nucleus, cytoplasmic expression is also slightly. vWFAb could significantly inhibit the proliferation ability of SW480 cells, and the effect was dose and time dependent(P<0.05); with 20 mg/L vWF antibody after 48 h, SW480 cells adhesion decreased significantly (P<0.05); Transmembrane migration of cells significantly decreased [(54.60±11.01) than (97.27±10.01) P<0.01].Conclusions Human colorectal cancer cells can express vWF; vWF in human colorectal cancer cells proliferation,adhesion and migration plays an important role in promoting. |
