Part oneObjective To investigate the expression of chemokine receptor CXCR4 and IL-6 proteins in colorectal carcinoma and determine their clinical significance through finding their association with MVD and correlation with each other.Methods Immunohistochemistry staining(SP method) was used to detect the expression of CXCR4, IL-6 proteins. CD34 immunostaining was used to examine the microvessel density.The correlation between the the expression of chemokine receptor CXCR4 and IL-6 and MVD was evaluated.Results The expression level of CXCR4 in colorectal cancer tissues was 72.34% (68/94), and the expression level of IL-6 in colorectal cancer tissues was 28.72% (27/94); The expression level of CXCR4 in para-cancerous mucosa was 9.677%(3/31), and the expression level of IL-6 in para-cancerous mucosa was 6.452%(2/31). The expression levels of CXCR4 and IL-6 in colorectal cancer tissues were significantly higher than that in para-cancerous mucosa. Expression of CXCR4 in colorectal carcinoma was correlated to invasive depth, lynph node metastasis(P<0. 05). Expression of IL-6 in colorectal carcinoma was not correlated to invasive depth, lynphnode metastasis or tumor locus of the colorectal carcinoma (P>0.05). CRCs containing overexpressed nuclear CXCR4 were significantly associated with invasive depth and lynph node metastasis.None of these clinicopathological factors were associated with the cytoplasmic CXCR4 expression. The expression of CXCR4 and IL-6 in colorectal carcinoma were correlated to MVD (P<0.05). The expression of CXCR4 was not correlated to the expression of IL-6 in colorectal carcinoma (r=0.4350, P>0.05).Conclusion The increased expression of CXCR4 and IL-6 in colorectal cancer tissue maybe relate with tumor angiogenesis and they may play a key role in colorectal cancer formation and metastasis. Part twoObjective To investigate the expression in human colon carcinoma cell line SW480 and effect on the capacity of proliferation.Methods Human colon carcinoma cell line SW480 cultured in vitro was treated with CXCR4 antibody. The expression of CXCR4 in SW480 was detected by ICH and IF. The effect of CXCR4 on the proliferation of human colon carcinoma cell line SW480 in vitro was explored by MTT colorimetric assay.Results DKK3 was expressed in the human colon carcinoma celll ine SW480; Compared with the control group, the proliferation ability of the SW480 cells decreased significantly after treatment with DKK3 antibody.Conclusion CXCR4 protein is expressed in colon cancer cell line SW480 cells, CXCR4 elevates the capacity of proliferation of colon cancer cell line SW480 in vitro.
