Objective : To observe the change of serum VE-cadherin endothelin-1(ET-1),nitric oxide(NO) and nitric oxide synthase(NOS) in newly diagnosed type 2 diabetic patients, and the correlaion among blood pressure(BP),body mass index(BMI),HbA1c,fasting blood glucose (FBG),2 hours postprandial blood glucose(P2hBG),cholesterin(CH), triglyeride(TG) , high density lipoprotein(HDL-c) , low density lipoprotein(LDL-c) ,Investigate the effect of insulin and gliclazide treatment on VE-cadherin,endothelin-1,nitric oxide ,nitric oxide synthase levels in newly diagnosed type 2 diabetic patients .Methods :Serum VE-cadherin, ET-1, NO and NOS levels were measured in thirty-five newly diagnosed type 2 diabetic patients,as well as BP,BMI,HbA1c,FBG, P2hBG,CH,TG,HDL-c,LDL-c,35 newly diagnosed type 2 diabetic patients were divided into two groups, including Gliclazide group 20 cases and insulin group 15 cases , Serum VE-cadherin, endothelin -1, nitric oxide and nitric oxide synthase levels were tested again after one-month treatment. Then compared with before treatment . 36 healthy subjects were selected as normal controls. Results :The level of VE-ca is higher in newly diagnosed type 2 diabetic patients ,so as to BMI,FBG,P2hBG,HbA1c,CH,TG,LDL-c, while NO is lower, The level of ET-1,NOS has not significant change. VE-cadherin was significantly positive correlated with HbA1c,while NO was negatively correlated with HbA1c. After gliclazide or insulin therapy the level of VE-Ca was obviously lower and the level of NO was obviously highe(rP<0.05). But after the two therapy, the levels of VE-ca and NO were no significant difference between them. Conclusion: The level of VE-ca is higher, while NO level is lower in newly diagnosed type 2 diabetic patients. It suggest that vascular endothelial function is injuried in newly diagnosed type 2 diabetic patients. After gliclazide or insulin therapy the level of VE-Ca was obviously lower and the level of NO was obviously higher , Which shows gliclazide and insulin has better protecting effect on vascular endothelial function, can improve VEC dysfunction. They effect the levels of VE-ca and NO but with no significant difference between them.
