The chemical constituents of Vitex trifolia L. and Prenanthes macrophylla Franch. have been studied in this thesis.Vitex trifolia L. is the species of Vitex genus belong Verbenaceae family. There are about 250 species distributed in tropical and temperate zone over the world, and 14 of these species distributed in our country. The investigation of pharmacology showed that they possess efficacy of analgesia,anti-inflammatory,anti-biosis,anti-tumor,inhibiting lymphocyte,hormonal regulation,anti-oxydation,ecdysis,helminthicide and so on. The chemical investigations of this genus indicated that ecdysterones,terpenoids,flavones,and lignanoids were its main components.Prenanthes macrophylla Franch. is the species of Prenanthes genus belong Compositae family. There are about 40 species distributed in tropical and temperate zone over the world,and 16 of these species distributed in our country. One species have long been used as folk medicine in China, possess efficacy of carbuncle and hemostasis. The chemical constituents of this genus have rare been reported, in previous investigation some triterpenes,sesquiterpenes and sterols have been isolated. The chemical constituents of these two plants have been studied. The compounds were isolated by means of silica gel,Sephadex LH-20 gel and MCI gel columm chromatography,HPLC etc. The structures of these compounds were elucidated by spectral methods (UV, IR, EI-MS, HR-ESI-MS, 1H-NMR, 13C-NMR, DEPT, 1H-1HCOSY, HSQC, HMBC, ROESY etc). 31 compounds are isolated from Vitex trifolia and 28 of them were identified as triterpenes,fatty acids,flavones and derivatives of benzene. Among them compounds V28 was a new natural product, 13 of them(V3,V4,V5,V7,V8,V11,V12,V15,V16,V18,V19,V24,V25) were first reported in Vitex genus. 11 compounds were isolated from Prenanthes macrophylla and 10 of them were identified as triterpenes,sesquiterpenes,flavones and organic acids.Among them one phenylacetyl glucoside was novel, all of the compounds are isolated from this plant for the first time. Moreover, some compounds were tested for their anti-tumor activity against several human tumor cell lines K-563, HL-60 and SMMC-7221 by standard MTT method. The result indicated that new compounds exihibited inactive, while two sesquiterpenes showed good anti-tumor activity. We developed a HPLC method to determine P4 in Prenanthes macrophylla Franch. and the good lingarity of this method was in the range of 0.44~3.52μg(r=0.999 7). This methods is simple and sensitive with good stability, it can be suitable for the determination of P4 in Prenanthes macrophylla Franch.The results in this thesis offer phytochemical proofs to plant chemotaxonomy and provide more chemical evidences for exploiting and utilizing the natural resources.In the end one review has been made for the bioactivities and chemical constituents of Vitex genus.
