5-Fluorouracil (5-Fu) is an anti-tumor drug through inhibiting cell cycles. It has been widely used in clinical application in recent years. However, in the process of clinical usage, 5-Fu behaves many defects because of its side effect and short half life. Drug delivery system could control the release, prolong the time of drug efficiency and transport drug to target-organ. Chitosan (CS) and Poly vinyl alcohol (PVA) are macromolecular to deliver drug and control the release of drug, due to their degradable properties and other superiorities.In this subject, 5-Fu/CS nanocompoud and 5-Fu/PVA nanocompoud are prepared by cryogenic process. The structure properties of drug-loaded nanocompoud are investigated by using scanning electron microscope (SEM). Nanocompoud shows stable and uniform superficial construction under the SEM. The larger the delivery concentration, the sticker the structure of nanocompoud, and the smaller the microchannel pore size. In the research of drug releasing capacity in vitro,5-Fu/CS nanocompoud quickly releases 5-Fu as to 56% in 2.6h,then slowly releases drug as to 78% in 48h,22% remaining.5-Fu/PVA nanocompoud quickly releases 5-Fu as to 46% in 2.6h,then slowly releases drug as to 70% in 24h,30% remaining. Meanwhile, we explore three factors that affect the percentage of cumulative release: the delivery varieties, delivery concentration, and rate of cryogenic process.This subject also determines inhibitory of 5-Fu/CS nanocompoud and 5-Fu/PVA nanocompoud to DLD1 tumor cells by the method of MTT. The conclusion reveals 5-Fu can quickly kill the tumor cells in a short time, but loses efficiency as time prolongs.5-Fu/CS nanocompoud and 5-Fu/PVA nanocompoud persist to release drug to inhibit tumor cells after 24 hours, 48 hours, 72 hours. Thus, these drug-loaded nanocompoud control slow-release of 5-Fu.
