Objective:To investigate the expression of wnt signaling in placenta of preeclampsia and the relationship with.the disease.Methods:The semi-quantitative method of reverse transcription polymerase chain reaction (PT-PCR) was employed to investigate the mRNA expression levels of wntl, wnt2 and sfrp4 in placenta tissue of 20 cases of normal term pregnant women and 40 cases of women with preeclampsia (15 cases of women with mild preeclampsia and 25 cases of severe preeclampsia).Results:1.The expression of wntl mRNA in placenta was significantly lower in the preeclampsia group (0.82±0.20) than the control one (1.40±0.27; P <0.05).The severe preeclampsia group(0.79±0.17) and mild preeclampsia group (0.86±0.22) were significantly lower compared with the control one (P<0.05). And the expression was lower in severe group than in the mild one, but there was no significant difference between them (P>0.05).2.The expression of wnt2mRNA in placenta was significantly lower in the preeclampsia group (0.39±0.20) than the control one (1.21±0.25; P<0.05).The severe preeclampsia group(0.27±0.09) and mild preeclampsia group (0.60±0.14) were significantly lower compared with the control one (P<0.05). And the expression was significantly lower in severe group than in the mild one (P<0.05).3.There was no significant difference of the sfrp4mRNA expression in placenta between preeclampsia group (0.80±0.19) and the control one (0.72±0.14;P>0.05). There was an increasing trend of sfrp4mRNA expression between the control group, the mild group(0.78±0.15) and the severe one (0.81±0.22), but there was no singnificant difference between them. 4.There was significant positive correlation between the level of wntlmRNA and wnt2mRNA expression in placenta of preeclampsia group (r=0.417, P<0.05), and negative correlation between the level of wnt1mRNA and sfrp4mRNA expression (r=-0.425, P<0.05),but there was no correlation between wnt2mRNA and sfrp4mRNA expression (r=-0.029,P>0.05).Conclusion:1.wnt1mRNA, wnt2mRNA and sfrp4mRNA are all expressed in placenta tissue of normal term pregnant women and women with preeclampsia.2.The decrease of the wntlmRNA and wnt2mRNA expression in placenta may participate in the pathogenesis of preeclampsia, and the expression of wnt2mRNA may have a relationship with its severity also. Further investigation of the relationship between the sfrp4mRNA and preeclampsia is required.3.wntl and wnt2 signaling may have some cross-talk in transmission, which may have combined action on the progress of preeclampsia. sfrp4 may play a role in restraining the wntl signaling, but have no same effect on wnt2 signaling.
