| PURPOUS Analysis the pertinence between the genomic polymorphism of two metabolic enzymes "CYP1A1" & "GSTM1" and the Genetic Predisposition of Teratospermia which caused by smokingMETHOD A group of 60 teratospermia specimens of sterile patients who were already approved had Teratospermia was collected in "the Second Xiangya Hospital of Central South University" from 2010/3/1 to 2010/5/1; Meanwhile, another so called "comparison group" of 60 specimens of sterile patients whose sperms were approved as natural was collected too. Use PCR-RFLP to examine the distribution frequency of genomic polymorphism of CYP1A1 & GSTM1 of both "teratospermia group" and "comparison group", analyze the pertinence of the two metabolic enzymes and teratospermia, and, the interaction between two metabolic enzymes ("CYP1A1" & "GSTM1") and the Genetic Predisposition of Teratospermia which caused by smoking.RESULTS1. Distribution frequency of CYP1A1 "teratospermia group" and "comparison group" is wild type 43.3%, hybrid type 23.3%, mutant 33.3%and 43.3%, genotype 43.3% and 11.7%, the comparison of frequency differences of two groups has statistical difference (X2=9.88,P=0.007); Distribution frequency of GSTM1 "teratospermia group" and "comparison group" GSTM1 (+)和GSTM1 (-) is 30%,70% & 53.3%,46.7%, the comparison of frequency differences of two groups has significant statistical differences (X2=6.72,P=0.01)2. Take CYP1A1 wild type as reference, CYP1A1 heterozygous genotype OR=0.48 (95%CI=0.20-1.15), no significant statistical differences (P=0.95); CYP1A1 mutant type gene OR=2.97 (95%CI=1.08-8.19), has significant statistical differences (P=0.03); Combined CYP1A1 heterozygous genotype and CYP1A1 mutant gene, the dangerous degree has increased, but it has no significant statistical differences (OR=1.07,95%CI=0.52-2.20, P=0.85)3. Take GSTM1 (+) as reference, GSTM1 (-) OR=2.667, 95%CI=1.26-5.64, has significant statistical differences (P=0.01)4. Take CYP1A1 wild type from non-smoking person as reference, a non-smoking person who carries CYP1A1 heterozygous genotype doesn't have more danger to get teratospermia, a non-smoking person who carries CYP1A1 mutant type gene has more danger to get teratospermia, but it has no significant statistical differences.The dangerous degree in a smoking person who carries CYP1A1 wild type gene was 3.5 times as much as that (P=0.04); The dangerous degree in a smoking person who carries CYP1A1 heterozygous genotype was 2.4 times as much as that (P=0.19); CYP1A1 mutant gene was 5.3 times (P=0.02); Combined CYP 1A1 eterozygous genotype and CYP1A1 mutant gene was 3.2 times (P=0.54)。5. Take GSTM1(+),CYP1A1 wild type as reference. The dangerous degree in a person who carries GSTM1(+) and CYP1A1 mutant type gene was 1.24 times as much as that (95%CI=0.23-6.62), no significant statistical differences (P=0.8), But the dangerous degree in a person who carries GSTM1(-) and CYP1A1 mutant type gene was 3.71 times as much as that (95%CI=0.98-14.05), has significant statistical differences (P=0.03)。CONCLUSION1. Take a normal person to get teratospermia as reference, The dangerous degree in a person who carries CYP1A1 mutant gene has increased.2. Take a normal person to get teratospermia as reference, The dangerous degree in a person who carries GSTM1 (-) has increased significantly.3. Take a normal person to get teratospermia as reference, The dangerous degree in a person who carries both GSTM1 (-) and CYP1A1 mutant gene has increased significantly. |
