Objective To investigate the effect of Liposomal Quercertin on Advanced Glycation End Products(AGEs) and Receptor for Advanced Glycation End Products (RAGE) in retina of diabetic rats and its mechanisms.Methods Nanoliposomal Quercetin was prepared by thin-film evaporator and high pressure homogenization method. Experimental diabetic rats induced by streptozotocin (STZ) were randomly divided into the following groups:Diabetes Mellitus (DM) control group(without treatment); Liposomal Quercetin low, middle and high doses groups (treated with Liposomal Quercetin by intragastric administration respectively using 50mg/kg.d,150mg/kg.d and 250mg/kg.d); aminoguanidine control group (treated with aminoguanidine by intragastric administration using 100mg/kg.d). After 12 weeks, the morphological changes changes of retinal was observed by hematoxylin-eosin staining, and the expression of AGEs and RAGE mRNA in retina were detected through immunofluorohistochemistry and reverse transcription polymerase chain reaction (RT-PCR), and the data were compared with normal control group.Results1. Retinal:Remarkable pathological changes of retinal of diabetic rats were found in Diabetes mellitus control group. In Liposomal Quercetin middle doses group, the pathological changes of retinal of diabetic rats were reduced.2. The body weight in Liposomal Quercetin low, middle and high doses groups were similar to that in diabetes mellitus control group; The blood glucose in Liposomal Quercetin low, middle and high doses groups were lower than that in diabetes mellitus control group, but there was no statistical significance.3. The level of AGEs in Liposomal Quercetin groups and aminoguanidine group was lower than that of diabetes mellitus control group (AGEs:537.29±103.21). The level of RAGE mRNA (1.192±0.098) in Liposomal Quercetin groups and aminoguanidine group was lower than that of diabetes mellitus control group, and the Liposomal Quercetin middle dose group (AGEs:478.75±90.34,RAGE mRNA:0.724±0.028) decreased significantly (P<0.01)Conclusion 1. Liposomal Quercetin could reduce the change of pathomorphology in retina and improve the general condition of diabetic rats induced by streptozocin to some extent, and there may have a hypoglycemic effect. The above conclusion needs to be further proved.2. Liposomal Quercetin can intervene the development of retinal microangiopathy in early period by reducing the level of AGEs and RAGE mRNA in retina diabetic rats effectively.3. This study provides a basis for the clinical applications of Liposomal Quercetin, and provides new ideas and theories for treatment of DM in Traditional Chinese.
