Objective:Observation of calcium alginate/RGD and chondrocytes, osteoblasts composite repair rabbit articular cartilage full-thickness defects of the short-term effects and feasibility. For the clinical application of cartilage tissue engineering methods to repair articular cartilage defects provide some experimental basis.Methods:Taking 1-week-old Japanese white rabbits inside the double-knee cartilage and calvaria bone, respectively, enzyme digestion method and flip dry put France isolated primary chondrocytes and osteoblasts, and then subcultured, and have enough cartilage cells and bone cells, and RGD and sodium alginate mixed use adult Japanese white rabbits 36, then divided into A, B, C three groups:A:test group; B:control group;C:blank group. Respectively, both sides of the femoral intercondylar fossa caused by full-thickness cartilage defect model, and then do the appropriate processing:A group implanted calcium alginate/cell complex+RGD; B group implanted calcium alginate/cell complex; C group of defects exclusion. Conventional farming, respectively, drawn after 4,8,12 weeks to observe the repair of cartilage defects, and its evaluation.Results:Animal experiments showed that the experimental group in a transparent cartilage repair cartilage defects mainly; control group, consisting mainly of a transparent cartilage repair, but the quality of cartilage was significantly lower than the test group; blank group mainly filled with fibrous tissue.Conelusion:RGD can significantly promote cell adhesion,accordingly,to better maintain the cell phenotype; Application of cartilage tissue engineering methods to repair cartilage defects in the short-term result is good, it can better repair articular cartilage defects, restore joint shape and function.
