BCG-PPD Intravitreal Injection Of Purified Protein To Induced Animal Model Of Eales Disease

Objective To analyze the BCG purified protein derivative intravitreal injection to establishing an animal model of Eales Disease. Methods Adult male SD rats(N=96) were randomly divided into four groups:Sensitizedâ… (N'=24),â…¡(N'=24),â…¢(N'=24)and non-sensitized group(N'=24),each of which further divided into four sub-groups(n=6).Sen-sitized groupâ… :sensitized with BCG-PPD5U+FICA;Sensitized groupâ…¡:concentration of lmg/ml of HS-AgP35+FICA mixed in equal volumes of adjuvant breast study uniform emulsion bowl,take0.2ml antigen injected into the foot pad skin; Sensitized groupâ…¢:The concentration of lmg/ml of HS-AgP35+FICA equal volume of adjuvant breast study uniform emulsion bowl,take 0.1ml antigen injecte into one foot pad skin of rats,the other side of the foot pad with BCG-PPD 5U+FICA line of intradermal injection of allergens;non-sensitized group received no treatment.14d later,intravitreal injectionBCG-PPD5U,10U,20Uandsaline.Through fluorescein fundus angiography(FFA),retina,retinal pathology,serum anti-HS-AgP35 antibody, anti-dsDNA antibodis in serum,MTT and other indicators of the examination and testing to make related analysis.Resrults 1.determination of the anti-HS-AgP35 antibodies in blood serum:1.1 compared to the anti-HS-AgP35 antibodies in different doses of the same group in the blood serum:In the Sensitized groupâ… :the concentration of anti-HS-AgP35 antibody of 5U group in serum was higher than NS group,10U group, each two groups had significant difference (P=0.005, 95%CI0.0158-0.0732;P=0.002,95%CI0.0211-0.0739); In the Sensitized groupâ…¡:the concentration of anti-HS-AgP35 antibody of the 20U group and 10U group in the serum were higher than the 5U group, each two groups had significant diference (P=0.022, 95%CI0.0661-0.0705;P=0.003,95%CI0.0200-0.0844); the concentration of anti-HS-AgP35 antibody between the Non-sensitized group and the Sensitized groupâ…¢had no significant difference(P>0.05).1.2 compared to the anti-HS-AgP35 antibodies in different group of the same dose in the blood serum:5Ugroup:the concentration of anti-HS-AgP35 of the sensitized groupâ… andâ…¢in the serum were higher than the Sensitized groupâ…¡,each two groups had significant difference(P=0.000,95%CI0.0444-0.0783;P=0.045,95%CI0.0017-0.1243);10U group:the concentration of anti-HS-AgP35 of the sensitized groupâ…¡orâ…¢in serum were higher than the non-Sensitized group and the Sensitized groupâ… ,each two groups had significant difference(P=0.007,95%CI0.0088-0.0482;P=0.017,95%CI0.0048-0.0442;P=0.002,95%CI0.0133-0.0527;P=0.006,95%CI 0.0093-0.0487).2.determination of the anti-dsDNA andibodies in blood serum:2.1. compared to the anti-dsDNA andibodies in different doses of the same group in the blood serum:sensitized groupâ…¡:the concentration of anti-dsDNA antibodies of the 10U group in serum were hegher than the NS group,5U group and 20Ugroup,each two groups had significant diference(P=0.003,95%CI0.0164-0.0680; P=0.009,95%CI0.0097-0.0613; P=0.004,95%CI0.0145-0.0661). the Non-sensitized group,the Sensitized groupâ… andâ…¢each two groups had no significant difference(P>0.05).2.2 compared to the anti-dsDNA andibodies in different group of the same dose in the blood serum:the 5U group:the concentration of anti-dsDNA antibodies of the Sensitized groupâ… in serum was higher than the Non-sensitized group and the Sensitized groupâ…¢,each two each two groups had significant diference (P=0.021,95%CI 0.0057-0.0626;P=0.008,95%CI0.0115-0.0685).the 10U group:the concentration of anti-dsDNA antibodies of the Sensitized groupâ…¢in serum was higher than the Non-sensitized group and the Sensitized groupâ… , each two groups had significant difference (P=0.016,95%CI 0.0152-0.1295;P=0.010,95%CI 0.0210-0.1353). the 20U group:the concentration of anti-dsDNA antibodies of the Sensitized groupâ…¢in serum was higher than Non-sensitized group,two group had significant diference (P=0.022, 95%CI 0.0041-0.0475). the NS group:the Non-sensitized group and the Sensitized group had no significant difference(P>0.05).3.Lymphocyte transformation test:the lymphocyte stimulation index of serum in the sensitized groupâ…¢was higher than the Non-sensitized group and the Sensitized groupâ… , each two groups had significant diference (X2=9,X2=9,P=0.016,P=0.028), the lymphocyte stimulation index of serum in the sensitized groupâ…¡was higher than the Non-sensitized group, two groups had significant diference (X2=7,P=0.015).the Sensitized groupâ… compared with the Non-sensitized group or the Sensitized groupâ…¡,each two groups had no significant diference(P>0.05), the Sensitized groupâ…¡compared with groupâ…¢, two groups had no significant diference(P>0.05),4 FFA examination:infection different doses of BCG-PPD into vitreous space of the the Non-sensitized group and the Sensitized groupâ… ,two eyes occurs anterior segment inflammation,fundus fluorescein angiography(FFA) of ertinal vein dklatation,circuity,vein wall was stained,and no retinal hemorrhage point and exsudation,no blocked fluorescence. Sensitized groupâ…¡occurs fluorescence liakage. the sensitized groupâ…¢occurs fluorescence liakage and see images of retinal neovascularization. four groups showed non-perfusion area,no exudative retinal detachment.5.Retinal pathology and FITC-dextran angiography:Retinal pathology show the retina of non-sensitized group and the sensitized groupâ… , thickening,fold,structural disorder,retinal vascular dilation,congestion,no perivascular lymphocytic inliltration,and no veovascularization. Sensitized groupâ…¡groupâ…¢addition to the above,also found beside a small amount of retinal vascular lymphocytic infiltration.FITC-dextran angiography show non-sensitized group of vascular disorder of moving,the wall was stained,blood vessels nodular protrusions,no fluorescent liakage;sensitized groupâ… can be seen the edge of FITC-dextran fluorescence liakage;stenosis occurred in sensitized groupâ…¡;sensitized groupâ…¢see vascular occlusion and non-perfusion area. Conclusion 1.after foot pad of rats intradermal sensitization with HS-AgP35 united BCG-PPD antigen,intravitreal injection of BCG-PPD can produce inflammation of retinal blood vessels;2.intravitreal injcetion of BCG-PPD,the united sensitized group is more easy than the single sensitized and non-sensitized group to produce retinal vascular inflammation,intravitreal injcetion of different concentrations of BCG-PPD,which induced the immune response and the extent of retinal vascular inflammation differ;3.speculated that the vitreous through the injection of the BCG-PPD to the sensitized rats,the body may be caused retinal vasculitis by sensitized T/B lymphocyte.
